甲状旁腺素及其受体对成釉细胞增殖的影响

摘要 目的通过体外培养成釉细胞,探讨甲状旁腺素(PTH)对于小鼠成釉细胞增殖的影响。方法首先利用免疫组化方法检测甲状旁腺素1受体(PTH1R)在小鼠牙齿成釉细胞中的表达;然后在细胞培养箱内培养成釉细胞16h,通过利用不同浓度的PTH作用于成釉细胞,噻唑蓝荧光检测PTH对成釉细胞的增殖作用效果。结果免疫组化法检测可知PTH1R在小鼠成釉细胞层中表达呈现阳性;荧光检测发现PTH可显著促进成釉细胞的增殖,尤其是在浓度达到1μg/L时其促进细胞增殖效果达到最高值。结论 PTH1R在小鼠牙齿成釉细胞层中具有较强的表达效应,同时PTH可以促进成釉细胞的增殖,其有效作用浓度与成釉细胞生长密度呈相关关系。 Objective To study the regulatory effects of PTH on proliferation of ameloblasts in vitro .Meth-ods PTH1R expressed in ameloblasts of mouse teeth was examined by immunohistochemical technique .The ameloblasts were cultured for 16 h in incubator ,trypan-blue fluorescence detection methods was used to evaluate the effects of PTH on ameloblasts proliferation in different concentrations .Results Positive expression of PTH1R was observed in the mouse ameloblasts layer by immunohistochemical technique .Fluorescence detection showed that PTH could significantly pro-mote the proliferation of ameloblastat ,when the concentration of PTH reach 1μg/L,the promote effect of ameloblasts pro-liferation could reached the highest value .Conclusion PTH1R has strong expression effect in mouse teeth ameloblasts layer .Such findings may be important in the response of these cells after tissue injury .PTH can stimulate the proliferation of ameloblasts .
