胰性脑病大鼠血脑屏障通透性改变的机制研究 被引量:5

摘要 目的探讨胰性脑病(Pancreatic Encephalopathy,PE)大鼠血脑屏障(Blood brain Barrier,BBB)通透性变化及其意义。方法 40只Sprague-Dawley(SD)大鼠,随机分成假手术(Sham-operation,SO)组和PE组。逆行胰胆管注射5%的牛黄胆酸钠构建PE模型。伊文思蓝(Evans Blue,EB)测定血脑屏障通透性;HE染色观察胰腺和脑组织病理变化。Elisa法检测脑组织中TNF-α和LI-1含量,比色法检测血清SOD和MDA含量。结果 PE大鼠脑内EB含量及其胰腺和脑病理评分均升高,血清SOD下降而MDA则升高,脑组织中TNF-α和IL-1含量升高,与SO组比较有统计学意义;相关分析发现脑内EB含量与脑组织病理评分及TNF-α和IL-1含量均呈正相关。结论 PE大鼠的BBB通透性升高并与脑损伤正相关,我们推测氧化应激和炎症反应参与破坏BBB的完整性从而诱发PE发病。 Objective To explore the significance and permeability change of blood brain barrier( BBB)of rat with pancreatic encephalopathy( PE). Methods Forty Sprague- Dawley( SD) rats were randomly divided into sham operation( SO) group and PE group. PE rats were induced by retrograde duct infusion of 5%sodium taurocholate( STC). The permeability of BBB was tested by Evans Blue,the pathological changes of pancreas and brain were observed by hematoxylin and eosin stain. The contents of TNF- α and interleukin- 10( IL- 10) in brain were tested by enzyme- linked immunospecific assay( ELISA). The contents of superoxide dismutase( SOD) and malondialdehyde( MDA) in brain were determined by chromatometry. Results The contents of EB and pathological scores of brain and pancreas in PE rats were increased,the contents of TNF- αand IL- 10 in brain of rats with PE were enhanced obviously,the level of MDA in serum was up- regulated,but SOD was decreased,the differences were statistically significant when compared with SO group( P〈0. 05).There were positive correlation between EB contents in brain and brain pathological scores and the levels of inflammatory factors( TNF- α,IL- 1beta) in brain of PE rats. Conclusions The BBB permeability of PE rats has a positive relationship( correlation) with brain injury. Therefore,we presume that oxidative stress and inflammatory reaction might be involved in BBB integrity damage.
