苦瓜蛋白MAP30的抗菌作用及PTD-MAP30跨克氏原螯虾肠膜功能的研究 被引量:3

摘要 根据Gen Bank上MAP30基因的全序列,设计引物PCR扩增得到MAP30基因,并通过重叠PCR方法扩增得到PTD-MAP30基因,构建重组表达载体MAP30-p ET28a(+)和PTD-MAP30-p ET28a(+),转化大肠杆菌BL21(DE3)。基因工程菌株在18℃ IPTG条件下诱导,表达产物经SDS-PAGE和Western blot检测显示有与预期大小相符合的蛋白,并用Ni2+-柱纯化目的蛋白。抑菌圈实验发现MAP30蛋白对弧菌敏感性较高,MTT法测得最小抑制浓度MIC为300μg/m L,生长抑制曲线显示蛋白对弧菌的抑制作用具有浓度依赖性。ELISA实验说明TAT-MAP30蛋白可以快速穿过克氏原螯虾的肠上皮细胞并进入到血淋巴中,在1 h左右检测到的蛋白量最多,而且维持时间可以达到6 h。
