摘要 目的:探讨维生素E对锰中毒小鼠学习记忆及海马区caspase-3表达的影响。方法:将60只雄性昆明小鼠随机分为四组:对照组(CG)、维生素E组(VG)、锰中毒组(MG)、锰中毒加维生素E组(MVG),每组15只。采用腹腔注射氯化锰的方式制作锰中毒模型,Morris水迷宫观察小鼠的学习记忆能力,免疫组织化学方法检测caspase-3蛋白的表达。结果:Morris水迷宫定位航行实验显示,CG、VG、MVG三组的逃避潜伏期比MG组明显缩短,空间探索实验中,CG、VG、MVG三组穿越平台的次数明显多于MG组。免疫组织化学实验中,MG组海马CA1区和DG区caspase-3的表达明显多于CG组,MVG组海马区caspase-3的阳性细胞数明显少于MG组。结论:维生素E干预可减少海马区神经细胞的凋亡,改善锰中毒小鼠的学习记忆能力。 Objective: To study the effect of vitamin E on learning and memory and the expression of hippocampus caspase-3 in manganese poisoned mice. Methods:Sixty male Kunming mice were divided into four groups at random: the control group( CG), the vitamin E group( VG), the manganese poisoned group(MG) and the manganese poisoning with vitamin E group( MVG), 15 for each group. The model of manganese poisoned mice was prepared by intraperitoneal in- jection of manganese chloride. The learning and memory ability of the mice were observed by Morris water maze, and the expression of caspase-3 was tested by immunohistochemical methods. Results: Morris water maze navigation experiment showed that the escape latency was significantly shorter and the times of passing through platform in space probe test were higher in CG, VG, and MVG groups than that in MG. group. Meanwhile, the expression of caspase-3 in MG group was significantly higher than that in the CG group, the positive cells of caspase-3 in hippocampus area of MCG group was sig- nificantly fewer than those in MG group. Conclusion : Vitamin E intervention can reduce the apoptosis of hippocampal neu- rons and improve the learning and memory ability in manganese poisoned mice.
