振动运动对雌性大鼠化疗相关性疲乏影响 被引量:4

Effect of vibration exercise on chemotherapy-related fatigue in female rats

摘要 目的探讨振动运动对化疗相关性疲乏的作用及可能机制。方法 60只雌性SD大鼠随机分为对照组和实验组。将实验组大鼠腹腔注射2.5 mg/kg盐酸表柔比星,累积剂量达15 mg/kg建立化疗疲乏模型后,随机分为模型组,低、中、高运动量组(15 Hz、5 min,25 Hz、15 min,35 Hz、45 min),每组10只;每周运动6 d,连续4周。末次运动前采用鼠尾悬挂实验测疲乏程度,末次运动结束后即刻采血,酶联免疫吸附试验测定血清白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNFα)、IL-6水平。结果与对照组比较,模型组大鼠悬尾不动时间[(197.40±23.74)s]延长(P<0.01),血清IL-1β、TNFα水平[分别为(748.17±34.71)、(66.46±0.98)pg/m L]升高(P<0.01);与模型组比较,中运动量组大鼠悬尾不动时间[(167.90±28.12)s]缩短(P<0.05),中、高运动量组大鼠血清IL-1β、TNFα水平[分别为(561.72±48.16)、(64.60±0.65)与(567.17±27.50)、(64.54±0.55)pg/m L]下降(P<0.01),低、中、高运动量组大鼠血清IL-6水平[分别为(89.07±3.22)、(118.40±5.10)、(130.69±3.87)pg/m L]升高(P<0.05)。结论振动运动可减轻雌性大鼠化疗相关性疲乏,其机制可能与下调炎症因子水平、上调抗炎因子水平有关。 Objective To investigate the effect of vibration exercise on chemotherapy related-fatigue and its possible mechanism in rats. Methods Sixty female Sprague-Dawley(SD) rats were randomly divided into a control and an experiment group. The rats in the experiment group received intraperitoneal injection of 2. 5 mg/kg epirubicin hydrochloride at a cumulative does of 15 mg/kg to establish a model of chemotherapy-related fatigue, and then were randomly divided into a model group, and low- (15 Hz for 5 minutes), medium- (25 Hz for 15 minutes), and high-exercise group (35 Hz for 45 minutes)( 10 rats in each group) and the vibration exercise was administrated to the rats 6 times a week for 4 weeks. Before the last exercise, the immobility time of tail suspension test(TST) were carried out to evaluate the status of fatigue. After the last exercise, all rats were sacrificed immediately, and enzyme-linked immunosorbent assay was employed to detect serum levels of interleukin-1 β ( IL-1 β ), tumor necrosis factor-alpha ( TNF-α ) and interleukin-6 (IL-6). Results The immobility time of TST(197.40 ±23.74 seconds) and the serum level of IL-1β(748. 17 234. 71 pg/mL) and TNFα (66.46 ± 0. 98 pg/mL)in model group were higher than those in the control group ( all P 〈 0. 01 ). The immobility time of TST( 167.90 ± 28. 12 seconds) in medium-exercise group was lower than that in the model group ( P 〈 0.05 ). The serum level of IL-1βα ( 561.72 ± 48.16 pg/mL and 64. 60 ± 0.65 pg/mL) and TNFα ( 567.17 ± 27.50 pg/mL and 64. 54 ±0. 55 pg/mL) of the rats in medium- and high-exercise groups were lower than those in the model group(both P 〈 0. 01 ). The serum level of IL-6 (89. 07 ± 3.22 pg/mL, 118.40 ± 5.10 pg/mL, and 130. 69 ± 3.87 pg/mL)of the rats in low-, medium-, and high-exercise groups were higher than that in the model group (all P 〈 0. 05). Conclusion Vibration exercise can reduce chemotherapy-related fatigue in female rats and the mechanism may associate with the down-regulation of inflammatory cytokines and up-regulation of anti-inflammatory cytokines.
