摘要
目的观察大黄提取物对高糖引起的心肌成纤维细胞增殖的影响并探讨其可能机制。方法心肌成纤维细胞随机分为正常组、高糖组和大黄提取物处理组(1、2.5、5、7.5、10、20、30和40μg/m L)。用cck-8(cell counting kit-8)试剂盒检测细胞活力,选择大黄提取物的最适浓度。Western blot法检测促凋亡蛋白bax、抗凋亡蛋白bcl-2、胶原蛋白Ⅰ以及cleaved-caspase3的表达。结果与正常对照组相比,高糖组的细胞活力增强,胶原蛋白Ⅰ、bcl-2表达明显增加,bax变化不明显;与高糖组相比,大黄提取物处理组胶原蛋白Ⅰ、bcl-2表达下调,bax和cleaved-caspase 3表达增加。结论大黄提取物可抑制高糖引起的成纤维细胞增殖和活化,其机制与激活线粒体凋亡通路有关。
Objective To observe the effect of rhubarb extract on the proliferation of myocardial fibroblasts induced by high glucose and to investigate its possible mechanism. Methods Myocardial fibroblasts were randomly divided into normal group, high glucose group and rhubarb extract treatment group(1、2.5、5、7.5、10、20、30 and 40 μg/m L). Cell viability was detected by cck-8(cell counting kit-8) reagent kit for selecting the suitable concentration of rhubarb extract. Western Blot assay was used to detect the expression of Bax, Bcl-2, collagen I and cleaved-caspase 3. Results Compared with the normal group, the cell activity in high glucose group was increased, the expression of collagen I and Bcl-2 was significantly increased, the expression of Bax was not obviously changed; Compared with the high glucose group, the expression of collagen I and Bcl-2 was down regulated, and the expression of Bax and cleaved-caspase 3 was increased in rhubarb extract group. Conclusion Rhubarb extract can inhibit the proliferation and activation of myocardial fibroblasts induced by high glucose, and its mechanism is related to the activation of the mitochondrial apoptotic pathway.
