微波预处理对油菜秸秆纤维素糖化的影响

摘要 [目的]考察微波预处理对油菜秸秆纤维素糖化的影响并优化处理工艺。[方法]以油菜秸秆为材料,采用单因素试验研究了微波处理功率、时间、液固比、p H及纤维素酶酶量、酶解时间等处理条件对产糖率的影响,并应用响应面分析法就预处理工艺进行优化,利用Design Expert 8.0.5b软件分析试验数据建立纤维素转化条件的二次项数学模型。[结果]试验表明,产糖率的模型均极显著(P<0.000 1),拟合度良好。微波预处理对油菜秸秆纤维素糖化的适宜工艺条件:微波功率560 W,微波时间7.00 min,液固比24.15∶1 ml/g,p H 5.95,酶量6 640 IU/g。在此条件下,产糖率为25.339 4%。[结论]研究可为油菜秸秆的综合利用提供技术支持。 [ Objective ] To investigate effects of microwave pretreatment on the cellulose saccharification of rape straw and optimize processing technique. [ Method ] The effects of treatment conditions such as microwave power, time, the ratio of substance to water, pH value, enzyme concentration and enzymatic hydrolysis time on the yield of sugar were studied by using single-factor test with rape straw as material. Then the interactional effects of microwave treatment time, ratio of substance to water, pH value and enzyme concentration on the yield of sugar were evaluated by using response surface method. Date were analyzed with Design Expert 8.0.5b software. [ Resuh] The results showed that good quadratic polynomial models of conditions of cellulose conversion were obtained, the model of yield of sugar was extremely significant ( P 〈 0. 000 1 ) and there were no lack of fit. The optimum conditions of microwave pretreatment on the cellulose saccharification of rape straw were as follows : microwave powder 560 W, microwave treatment time 7.00 rain, ratio of substance to water 24.15:1 ml/g, pH 5.95, enzyme con- centration 6 640 IU/g. Under these conditions, the yield of sugar was 25. 339 4%. [ Conclusion] The study can provide technical support for comprehensive utilization of rape straw.
