摘要
目的探讨非禁食和禁食对大鼠应激性胃溃疡胃黏膜损伤及MMP-9、TIMP-2表达的影响。方法非禁食应激组不予禁食,禁食应激组于应激前24 h禁食。用浸水-束缚应激(waterimmersion and restraint stress,WIRS)建立大鼠应激性溃疡模型。应激3 h后,观察非禁食应激组和禁食应激组大鼠胃黏膜病变,对溃疡指数(UI)进行评分;组织学观察胃出血;用Western blot检测胃黏膜MMP-9、TIMP-2的蛋白表达。结果 WIRS应激3 h能成功诱导大鼠应激性胃溃疡。非禁食应激组与禁食应激组比较,应激性胃溃疡大鼠的UI后者高于前者(P<0.01),黏膜层炎性细胞浸润数后者高于前者(P<0.01),WIRS大鼠胃黏膜MMP-9蛋白表达后者显著高于前者(P<0.01),而TIMP-2蛋白表达后者略低于前者(P>0.05)。结论非禁食应激可显著减少MMP-9表达。非禁食处理减轻浸水-束缚应激大鼠胃黏膜损伤。
Objective To investigative the influence of non-fasting and fasting treatments on water- immersion and restraint stress (WIRS)-induced gastric mucosal injury and on the expression of MMP-9 and TIMP-2 in rats. Methods The non-fasting group was not fasting, and the fasting group was fasting at 24 h before WIRS. Stress-induced ulcer model was established by WIRS. The gastric mucosa was examined and the ulcer index (UI) was evaluated after 3-hour WIRS. Mucosal injury was observed with histological method. In addition, the protein expressions of MMP-9 and TIMP-2 were measured by Western blotting. Results The rat ulcer model was established by 3-hour WIRS. The UI, inflammatory cells and MMP-9 protein expression in the mucosa were higher in the fasting group than in the non-fasting group ( P 〈 0. 01 ), while the TIMP-2 protein expression was slightly lower in the fasting group (P 〉 0. 05 ). Conclusion alleviate WIRS-induced gastric mucosal injury through significantly down-regulating in the rats. Non-fasting treatment can the expression of MMP-9 in the rats.
