祛风通络方对阿霉素大鼠肾小球内皮细胞VEGF及其受体FLK-1表达的影响

Qufengtongluo recipe on expression of VEGF and its receptor FLK-1 in adriamycin-induced rat glomerular endothelial cell

目的研究祛风通络方对阿霉素模型大鼠肾小球内皮细胞VEGF及其受体FLK-1表达的影响。方法130只SD大鼠抽出30只作为空白对照组(A组),余下大鼠采用阿霉素给予尾静脉一次性注射构建大鼠肾病模型,造模成功后,随机分为模型组(B组,n=18)、祛风通络方组(C组,n=18),祛激组(祛风通络方和激素共用组,D组,n=18),激素组(E组,n=18),苯那普利组(F组,n=18),构建肾小球内皮细胞阿霉素模型,给予相应大鼠血清治疗,邻苯三酚红/钼酸盐法检测24 h尿蛋白定量;全自动生化分析仪检测血清白蛋白(ALB)、血脂(TG)、肾功能BUN、Cr;Western Bloting及RT-PCR检测肾小球内皮细胞VEGF及其受体FLK-1的表达。结果 C、E组能升高大鼠血清ALB的含量,降低血清TCH的含量,与B组比较,差异均有统计学意义(P<0.01);肾小球内皮细胞经阿霉素损伤后,B组较A组VEGF、FLK-1蛋白及mRNA的表达均升高,差异有统计学意义(P<0.01),各组加入相应血清冻干粉后,C、E组VEGF、FLK-1蛋白表达均下调,与B组比较,差异有统计学意义(P<0.01)。结论祛风通络方能够下调肾小球内皮细胞VEGF及其受体FLK-1的表达,改善阿霉素大鼠肾病模型的肾功能。

Objective To study the effects of qufengtongluo recipe on the expression of VEGF and its receptor FLK-1in adriamycin-induced rat glomerular endothelial cell. Methods The 30 out of 130 SD rats are used as the control group（ A）,and the remaining rats are given intravenously adriamycin one-time injecting to construct nephropathy model,and after the model is successfully constructed,rats are randomly divided into model group（ B,n = 18）,qufengtongluo eecipe group（ C group,n = 18）,qufeng and hormone group（ qufengtongluo recipe and hormones share group,D,n = 18）,hormone group（ E,n = 18）,benazepril group（ F,n = 18）,to build adriamycin-induced glomerular endothelial cells models,and to give the appropriate serum for treatment,and pyrogallol red/molybdate method is used to detect 24 h urinary protein; automatic biochemical analyzer is used to detect serum albumin（ ALB）,lipids（ total cholesterol,TCH; Triglycerides,TG）,renal function（ blood urea nitrogen,BUN; serum creatinine,Cr） and RT-PCR;western bloting and RT-PCR are used to detect the expression of VEGF and FLK-1 in the cells. Results Both C,E group can elevate serum ALB,decrease serum TCH,and the differences are significant comparing with group B（ P〈0. 01）; after glomerular endothelial cells are damaged by ADR,the expression of VEGF and FLK-1proteinand mRNA in group B is higher than that in group A,the difference is significant（ P〈0. 01）,and after adding the appropriate serum lyophilized powder,the expression of VEGF and FLK-1 protein and mRNA in group C,E are down-regulated compared with group B（ P〈0. 01）. Conclusion Qufengtongluo recipe can decrease expression of VEGF and its receptor FLK-1 in glomerular endothelial cells,and improve renal function in rats with Adriamycin-induced nephropathy model.
