冬虫夏草浸出液合用硫唑嘌呤对大鼠肝移植后免疫调节的影响 被引量:5

摘要 目的观察冬虫夏草浸出液合用硫唑嘌呤对大鼠减体积1/2肝脏移植后免疫调节功能的影响。方法80只大鼠,随机分为空白对照组、模型组、硫唑嘌呤组、冬虫夏草组,采用改良双袖套法进行1/2大鼠同种异体减体积原位肝脏移植,术后硫唑嘌呤组用硫唑嘌呤稀释溶液灌胃1.0 mg/(kg·d)、冬虫夏草组用6%冬虫夏草浸出液(10 m L/kg)加硫唑嘌呤1.0 mg/(kg·d)灌胃。于治疗21 d后,测定肝微粒体细胞色素P450亚型酶(CYPIA2、CYP2C9、CYP2D6、CYP2E1、CYP3A4)活性,血清总蛋白、白蛋白、血红蛋白,静脉血红细胞、白细胞、T淋巴细胞、单核细胞数量,受体大鼠感染率。结果冬虫夏草浸出液明显抑制存活受体大鼠P450亚型酶CYPIA2(0.68±0.09)、CYP3A4酶的活性(4.92±0.27),降低静脉血红细胞(6.71±0.87)、白细胞数量(10.36±0.09),提高血清总蛋白(19.39±4.93)、白蛋白(14.27±4.45)及血红蛋白含量(19.39±4.973),明显降低受体大鼠感染率40.87%。与硫唑嘌呤组比较,P<0.05。对T淋巴细胞及单核细胞数量无明显影响,与硫唑嘌呤组比较,P>0.05。结论冬虫夏草浸出液在不影响硫唑嘌呤抑制肝移植排斥反应的同时,改善肝脏移植后低蛋白血症,提高体液免疫,对细胞免疫功能无明显影响,具有双向免疫调节作用。 Objective To observe the effects of cordyceps sinensis extract combined with azathioprine on rat reduced size liver 1 /2 immune function after transplantation. Methods 80 rats are randomly divided into blank control group,model group,azathioprine,cordyceps sinensis group,1 /2 of rat orthotopic liver transplantation with reduced size modified cuff technique,postoperative azathioprine azathioprine dilute solution by gavage 1. 0 mg /( kg · d),Cordyceps sinensis extract group 6%( 10 mL/kg) + oral azathioprine 1. 0 mg /( kg·d) by gavage. In 21 days of treatment( d),the determination of hepatic microsomal cytochrome P450isoforms( CYPIA2,CYP2C9,CYP2D6,CYP2E1,CYP3A4)activity,serum total protein,albumin,hemoglobin,red blood cells,white blood cells,T lymphocytes,the number of mononuclear cells,receptor of rat infection rate. Results Cordyceps sinensis extract significantly inhibites the survival rate of the recipient rat P450 isoforms of CYPIA2( 0. 68 ± 0. 09),the enzyme activity of CYP3A4( 4. 92 ± 0. 27),lower vein red blood cells( 6. 71 ± 0. 87),the number of white blood cells( 10. 36 ± 0. 09),increased serum total protein,albumin( 19. 39 ± 4. 93)( 14. 27 ± 4. 45) and hemoglobincontent( 19. 39 ± 4. 973) is significantly lower in rats,the infection rate of 40. 87%. Compared with azathioprine( P〈0. 05),the differences are statistically significant. There is no significant effect on the number of T lymphocytes and mononuclear cells,compared with azathioprine group,P 〉0.05. Conclusion Cordyceps sinensis extract on azathioprine suppresses hepatic allograft rejection and at the same time,improve hypoproteinemia after liver transplantation,enhance humoral immunity,and has no significant effect on the cellular immune function,it has a dual immunomodulatory activity.
