一株拮抗椰子茎泻血病菌的链霉菌鉴定 被引量:4

Identification of a Streptomyces Strain Against Coconut Stem Bleeding Disease Caused by Ceratocystis Paradoxa

摘要 从文昌椰子园土壤分离筛选到一株对椰子泻血病菌奇异长喙壳菌(Ceratocystis paradoxa)有良好抑制效果的放线菌FXJ-258,对该菌株进行了形态特征、培养特征、生理生化特征以及16S r DNA序列分析。结果表明:该菌孢子方形至长圆形,表面光滑,在高氏培养基上,气丝初期白色,随着时间渐变为浅灰色,基丝无鉴别色素,无可溶性色素;能利用L-阿拉伯糖,D-半乳糖,D-葡萄糖,D-甘露糖,D-木糖,D-核糖,L-鼠李糖等碳源,不能利用D-果糖,肌醇,蔗糖,水杨苷等。以16S r DNA序列为基础构建了菌株FXJ-258的系统发育树,其与模式链霉菌株的16S r DNA序列的同源性为99.64%,初步将该菌株鉴定为Streptomyces murinus。 A streptomyces strain nam ed FXJ-258 against stem bleeding disease caused by Ceratocystis paradoxa was isolated from soil in Wenchang coconut garden. The morphological characteristics, cultural characteristics,physiological and biochemical characteristics, 16 S r DNA sequences analysis of Streptomyces FXJ-258 were studied.The result showed that, the spores of FXJ-258 were quadrate or ovale, smooth-surfaced. On Gause's Synthetic Agar medium(GSA), the substrate mycelia of FXJ-258 was colored that couldn't be identified, the clone of FXJ-258 was white when it started growing on GSA, after matured, the aerial mycelia of FXJ-258 became gray gradually, couldn't produce soluble pigment on GSA medium. FXJ-258 grew better on medium with L-arabinose,D-galactose, D-glucose, D-mannitol, D-xylose, D-ribose, L-rhamnose as C source; D-fructose, inositol, salicin and sucrose were not available. Besides, a phylogenetic tree was constructed by comparing with the published 16 S r DNA sequence of the related species; ITS sequence analysis revealed 99.64% sequence similarity with Streptomyces murinus. Thus, FXJ-258 was preliminarily classed to S. murinus.
