补阳还五汤对寒凝血瘀模型大鼠血液相关指标的影响 被引量:9

Effects of Buyang Huanwu Decoction on the blood related indicators in the cold stagnation and blood stasis rats

摘要 目的:从血液流变学和红细胞膜组分的变化探讨补阳还五汤改善寒凝血瘀模型大鼠血瘀证的作用机制。方法:40只Wistar大鼠,随机分为空白组、寒凝血瘀模型组、补阳还五汤高剂量组(14.0g/kg)、补阳还五汤低剂量组(3.5g/kg),每组10只。大鼠置于0-1℃冰水中8-10min,待大鼠全身僵直后取出,连续造模处理14d。从实验第1天开始,灌胃给药,1次/d。第15天大鼠皮下注射盐酸肾上腺素0.8m L/kg(0.8mg/kg),共2次,间隔4h,2次之间将大鼠置于冰水中浸泡5min,末次造模处理后禁食不禁水12h。取血测定血液流变学指标以及红细胞膜组分相关指标。结果:与模型组比较,补阳还五汤低剂量组能够显著降低全血黏度(P<0.05),升高红细胞变形指数(P<0.05,P<0.01),降低纤维原蛋白(FIB)(P<0.01)和血浆胆固醇(CHO)(P<0.05);补阳还五汤高剂量组可显著升高红细胞膜的超氧化物歧化酶(SOD)活力(P<0.05),补阳还五汤低剂量组可升高唾液酸含量(P<0.05)。结论:补阳还五汤可能通过降低血浆中FIB含量及血浆CHO含量,以及增加红细胞膜表面唾液酸和SOD含量,从而减轻自由基对红细胞攻击程度,提高抗氧化能力。 Objective:To investigate the mechanism of action in blood stasis due to cold accumulation by Buyang Huanwu Decoction(BYHWD).The change of hemorheology and the components of erythrocyte membrane were studied.Methods:Forty Wistar rats were randomly divided into blank group,model group and BYHWD high-dose group(14.0g/kg) and low-dose(3.5g/kg)group with 10 rats in each group.These rats were put into ice water(0- 1℃) for 8- 10 minutes until the rats were titanic everyday.The BYHWD were given by gavage everyday and this method was used for 14 days.In the 15 th day 0.8mL/kg adrenalin hydrochloride was injected into rats with hype for twice which interval 4 hours.The rats were put into ice water for 5 minutes during the interval.The related indexes of hemorheology and the components of erythrocyte membrane were determined.Results:Compared with the model group,the whole blood viscosity(P〈0.05),the content of cholesterol(CHO)(P〈0.05) and fibrinogen(FIB)(P〈0.01) in plasma were decreased by BYHWD.The red blood cell deformability(P〈0.05,P〈0.01),the content of sialic acid(P〈0.05) and SOD(P〈0.05) in erythrocyte membrane were increased by BYHWD.Conclusion:BYHWD could decrease the content of FIB and CHO in plasma.BYHWD also could improve the situation of blood stasis through increasing the content of sialic acid and SOD.It indicates that the ability of anti-oxidation is enhanced and free radical damaging is reduced to red blood cells.
