摘要
目的:研究祛浊通痹方(QZTB)抗炎镇痛及对脂多糖(LPS)诱导Raw264.7细胞的抗炎活性。方法:采用二甲苯致小鼠耳肿胀法评价QZTB抗炎作用以及热板法和醋酸致扭体法评价其镇痛作用。ELISA法测定LPS诱导Raw264.7细胞培养上清的前列腺素E2(PGE2)含量。结果:QZTB中、高剂量组能有效缓解小鼠二甲苯所致的耳廓肿胀(P<0.05,P<0.01),而QZTB低、中、高剂量组均明显提高小鼠痛阈值,并显著减少醋酸致小鼠扭体次数(P<0.01)。QZTB对LPS诱导Raw264.7细胞中PGE2的产生具有明显抑制作用(P<0.05,P<0.01)。结论:QZTB具有较好的抗炎镇痛作用及对体外LPS诱导Raw264.7炎性细胞具有抗炎活性,为进一步研究QZTB治疗痛风作用机制提供一定的理论依据。
Objective:To study the anti-inflammatory and analgesic effects of Quzhuo Tongbi Formula(QZTB) and its anti-inflammatory activities in Raw264.7 cells induced by lipopolysaccharide(LPS).Methods:Mouse models of ear swelling induced by dimethylbenzene were used to evaluate the anti-inflammatory action,and hot plate test and acetic acid twisting method were used to evaluate the analgesic effect.The concentration of PGE_2 in culture supernatants of Raw264.7 cells induced by LPS was tested by using ELISA.Results:QZTB could relieve the ear swelling of mouse induced by dimethylbenzene(P〈0.05,P〈0.01),improve the pain threshold of mouse,reduce the torsion times of mouse induced byacetic acid(P〈0.01),and inhibit the generation of PGE_2 in culture supernatants of Raw264.7 cells induced by LPS obviously(P〈0.05,P〈0.01).Conclusion:QZTB has obvious anti-inflammatory and analgesic effects and shows ananti-inflammatory activity in LPS-induced Raw264.7 cells in vitro,which provids a scientific theoretical basis for the further studies about the mechanism of treating gout with QZTB.
