异种雪旺细胞移植对大鼠损伤脊髓Nogo-A表达的影响 被引量:2

摘要 目的观察微囊化雪旺细胞对大鼠SCI后脊髓Nogo-A表达的影响,探讨微囊化异种雪旺细胞修复损伤脊髓的机制。方法通过对兔双侧坐骨神经干预变性处理后取出并进行细胞培养,通过使用海藻酸钡微囊化技术,制成微囊化雪旺细胞;实验大鼠随机分组:A组(微囊化雪旺细胞组);B组(雪旺细胞组);C组(单纯损伤组);D组(假手术组)。分组后取得脊髓损伤水平下脊髓组织进行Nogo-A免疫组化染色及Western-blot检测脊髓组织中Nogo-A蛋白表达水平。结果 SCI后A、B、C组大鼠右侧脊髓Nogo-A表达较D组明显增加。1周后A、B组Nogo-A表达较C组有显著下降(P<0.05),A、B组间Nogo-A表达无差异;SCI后4周,A组Nogo-A表达下调程度较B、C组更显著(P<0.05)。结论微囊化兔坐骨神经雪旺细胞移植可抑制脊髓损伤大鼠脊髓前角Nogo-A表达,促进轴突的再生,从而促进损伤神经元的修复。
