雷奈酸锶对糖尿病大鼠种植体骨结合的影响 被引量:1

Effect of strontium ranelate on implants osseointegration in diabetic rats

摘要 目的:评价口服雷奈酸锶对糖尿病大鼠种植体骨结合的影响。方法:20只成年Wistar大鼠采用高脂高糖饲料喂养+腹腔注射小剂量链脲佐菌素建立大鼠糖尿病模型,建模成功后,在大鼠股骨远端植入40枚钛种植体,每鼠2枚,术后实验组大鼠给予雷奈酸锶口服处理,对照组给予同等剂量的安慰剂,12周处死大鼠取材行显微CT扫描、组织学染色分析、最大拔出力检测,评价种植体骨结合效果。结果:通过高脂高糖特制饲料喂养8周后给予一次性腹腔注射小剂量链脲佐菌素,大鼠尾静脉取血检测血糖显著升高。术后12周取材发现与对照组相比,给予雷奈酸锶口服的大鼠骨体积分数(BV/TV),骨小梁厚度(Tb.Th)均显著高于对照组,骨小梁间隙(Tb.Sp)显著低于对照组。实验组与对照组骨结合率分别为(42.61±14.38)%,(35.57±9.74)%,最大拔出力分别为实验组(71.07±13.20)N,对照组为(55.36±19.68)N。结论:雷奈酸锶可促进糖尿病大鼠种植体骨结合。 Objective: To evaluate the effect of strontium ranelate on implants osseointegration in diabetic rats. Method: Diabetic rat model was established by diet enriched in fat and glucose and intraperitoneal injection of streptozotocin (STZ). Titanium implants were inserted into the distal femurs of the diabetic rats. Then the animals were divided into two groups: animals in experimental group were treated with orally strontiumranelate (625 mg / kg/day), the other animals received orally saline vehicle served as control group. Twelve weeks after implantation, all animals were sacrificed for micro-CT scanning, histological analysis and biochemical test. Result: After feed with diet enriched in fat and glucose and intraperitonealinjection of STZ, the fasting blood glucose were increased significantly. 12 weeks after implantation, the bone volume ratio (BV / TV) and trabecular thickness (Tb.Th) increased significantly in the experimental group, and compared to the control group, the trabecular space (Tb.Sp) decreased significantly. The bone implant contact (BIC) was (42.61±14.38)%, (35.57±9.74)% in experimental and control group respectively, and the maximum pull out force of experimental and control group were (71.07±13.20) N and (55.36±19.68)N. Conclusion: These results indicated the potential benefits of strontium ranelate under diabetic conditions to enhance osseointegration.
