摘要
目的:建立同时测定线纹香茶菜中咖啡酸、新西兰牡荆苷2、异夏佛塔苷含量的方法,以确定线纹香茶菜的最佳采摘期。方法:采用高效液相色谱法。色谱柱为Diamonsil C18,流动相为甲醇-0.1%磷酸溶液(梯度洗脱),流速为0.8 ml/min,检测波长为334 nm,柱温为25℃,进样量为15μl。结果:咖啡酸、新西兰牡荆苷2、异夏佛塔苷检测的质量浓度线性范围分别为0.23~5.68、0.31~7.76、0.45~11.30μg/ml(r=0.999 9、0.999 8、0.999 9);精密度、稳定性、重复性试验的RSD〈2.0%;加样回收率分别为95.1%~98.9%、97.4%~101.3%、95.5%~98.8%,RSD分别为1.8%、1.7%、1.4%(n=6)。采收于每年4-5月、7-8月的线纹香茶菜中上述3种成分含量最高。结论:该方法简便、准确、重复性好,可用于同时测定线纹香茶菜中咖啡酸、新西兰牡荆苷2、异夏佛塔苷的含量;线纹香茶菜的最佳采摘期为每年4-5月、7-8月。
OBJECTIVE: To establish a method for simultaneous determination of caffeic acid, vicenin-2 and isoschaftoside in Rabdosia lophanthoides to assess its optimal harvest period. METHODS: HPLC was performed on the column of Diamonsil C18 with mobile phase of methanol-0.1% phosphoric acid solution (gradient elution) at a flow rate of 0.8 ml/min, detection wave- length was 334 nm, column temperature was 25℃ and volume injection was 15 μl. RESULTS : The linear range was 0.23-5.68 μg/ml for caffeic acid(r=0.999 9), 0.31-7.76 μg/ml for vicenin-2(r=0.999 8) and 0.45-11.30μg/ml for isoschafloside(r=0.999 9); RSDs of precision, stability and reproducibility tests were lower than 2.0%, average recoveries were 95.1%-98.9% (RSD=1.8%, n=6), 97.4% -101.3% (RSD= 1.7%, n=6) and 95.5% -98.8% (RSD= 1.4%, n=6), respectively. The contents of above-men- tioned 3 ingredients of R. lophanthoides were the highest harvested in Apr. to May and Jul. to Aug. in a year. CONCLUSIONS: The method is simple, accurate and reproducible, and can be used for the simultaneous determination of caffeic acid, vicenin-2 and isoschaftoside in R. lophanthoides. The optimal harvest period is Apr. to May and Jul. to Aug. in a year.
