头顶一颗珠对肝癌小鼠免疫功能影响的实验研究 被引量:7

摘要 目的研究头顶一颗珠(TTM)注射液对肝癌模型小鼠免疫功能的影响。方法建立H22小鼠肝癌模型,将造模成功的肝癌小鼠依据随机数字表法分为模型组、氟尿嘧啶(5-FU)组、5-FU+TTM组和TTM组,每组10只。模型组、5-FU组每日按10 mg/kg腹腔注射生理盐水及5-FU,5-FU+TTM组在注射5-FU的同时每日腹腔按7.2 g/kg注射TTM,TTM组按7.2 g/kg注射TTM。给药5 d后测定各组胸腺、脾脏指数,检测外周血中白细胞、淋巴细胞及CD3、CD4、CD8细胞的百分比,血清中白细胞介素2(IL-2)、肿瘤坏死因子α(TNF-α)水平。结果 5-FU组和TTM组的胸腺指数与模型组比较差异无统计学意义(P>0.05),而5-FU+TTM组显著高于模型组(P<0.05)。与模型组比较,5-FU组外周血白细胞与淋巴细胞数量明显减少(P<0.05),TTM组显著增加(P<0.05);5-FU+TTM组白细胞数与模型组比较差异无统计学意义(P>0.05);与5-FU组比较,TTM组白细胞与淋巴细胞数量显著增加(P<0.05),5-FU+TTM组白细胞数量明显增多(P<0.05),但淋巴细胞数量变化差异无统计学意义(P>0.05)。相对于模型组,TTM组、5-FU+TTM组及5-FU组CD3、CD4百分比显著增加(P<0.05),CD8百分比变化差异无统计学意义(P>0.05);与较5-FU+TTM组比较,TTM组、5-FU组CD3、CD4百分比差异无统计学意义,但CD8百分比显著增加(P<0.05)。相对于模型组,TTM组、5-FU+TTM组及5-FU组IL-2、TNF-α水平显著增加(P<0.05);与TTM组比较,5-FU+TTM组、5-FU组IL-2水平差异无统计学意义(P>0.05),但5-FU组TNF-α水平显著增加(P<0.05),5-FU+TTM组差异则无统计学意义(P>0.05)。结论 TTM可以促进T细胞的成熟与迁移,提高肝癌小鼠IL-2、TNF-α水平,增强机体免疫力,具有抗肿瘤功能。 Objective To study the effect of Trillium tschonoskii Maxim( TTM) injection on immune functions of mice modeled with hepatoma. Methods H22 hepatoma-mice model was built, and the hepa-toma-mice with successful modeling were randomly divided into model group,5-FU group,5-FU+TTM group and TTM group,10 mice in each group. The model group mice and the 5-FU group mice were given physio-logical saline intraperitoneal injection with 10 mg/kg per day. The 5-FU+TTM group mice were given TTM intraperitoneal injection with 7. 2 g/kg at the same time, and the TTM group mice were injected with 7. 2 g/kg. The index of thymus and spleen,the proportion of leucocyte,lymphocyte and CD3 ,CD4 ,CD8 cells in the periphearl blood,and the concentration of IL-2,TNF-αin serum after five days′medicines were meas-ured. Results The thymus index of mice in 5-FU group and TTM group had no statistical significance and 5-FU+TTM group increased obviously compared with the model group(P〈0. 05);the amount of leucocyte and lymphocyte of peripheral blood in 5-FU group decreased obviously ( P 〈0. 05 ) and the TTM group increased obviously(P 〈0. 05)compared with the model group;the amount of leucocyte in 5-FU +TTM group had no statistical significance compared with the model group;the amount of leucocyte and lymphocyte in TTM group increased obviously compared with 5-FU group(P〈0. 05);the amount of leucocyte increased obviously in 5-FU+TTM group,but the amount of lymphocyte had no statistical significance. The proportion of CD3,CD4 in TTM group,5-FU+TTM group and 5-FU group increased significantly(P〈0. 05) and the proportion of CD8 had no statistical significance compared with the model group(P〉0. 05). The proportion of CD3 ,CD4 in TTM group,5-FU group had no statistical significance,but the proportion of CD8 increased obviously compared with 5-FU+TTM group(P〈0. 05). The contents of IL-2,TNF-αin TTM group,5-FU+TTM group and 5-FU group increased obviously compared with the model group(P〈0. 05);compared with TTM group,the contents of IL-2 had no obvious difference and had no statistical significance (P〉0. 05) in 5-FU+TTM group and 5-FU group,but contents of TNF-α in 5-FU group increased obviously(P〈0. 05) and the contents in 5-FU+TTM group had no statistical significance ( P 〉0. 05 ) . Conclusion TTM can advance the maturation and metastasis of T cells,improve the level of IL-2,TNF-α of mice with hepatoma, strengthen the organic immunity,thus has the effect of anti-tumor.
