血清白细胞介素18和环氧化酶2在大鼠脑缺血再灌注损伤过程中的水平变化

摘要 目的:观察白细胞介素18(interleukin-18,IL-18)和环氧化酶2(cyclooxygenase-2,COX2)在大鼠脑缺血再灌注损伤中的水平变化。方法:将40只成年健康SD大鼠随机分成5组,每组8只,假手术组、缺血组、再灌注2 h组(R2 h组)、再灌注12 h组(R12 h组)及再灌注24 h组(R24 h组)。构建大鼠脑缺血再灌注损伤模型,TTC染色,计算脑梗死体积,ELISA检测外周血IL-18和COX2的表达变化。结果:TTC染色显示,在缺血组和再灌注组都发现不同程度的脑梗死,缺血组梗死最严重,而假手术组未发现脑梗死。与假手术组相比较,缺血组和R2 h组IL-18和COX2水平明显升高,差异有统计学意义(P<0.05)。结论:大脑遭受缺血再灌注损伤时,炎症细胞因子IL-18和炎症介质COX2参与脑细胞死亡。 Objective: To investigate the levels of IL-18 and COX2 in rat cerebral ischemia-reperfusion injury. Methods: 40 healthy adult SD rats were randomly divided into 5 groups (n = 8), sham, ischemia, isehemia-2 hour reperfusion, ischemia-12 hour reperfusion, and ischemia-24 hour reperfusion groups. Rat cerebral ischemic-reperfusion injury model was induced by left middle cerebral artery occlusion. After TTC stain, the infarction volumes of the rat brain were calculated, and the levels of serum IL-18 and COX2 were examined by ELISA. Results: There were no any cerebral ischemia infarction in sham group, while the infarction lesions at different scales were observed in almost all experimental groups, with ischemia group the worst. In comparison to sham group, the levels of IL-18 and COX2 were obviously increased in both ischemia group and ischemia- reperfusion group (P 〈 0.05). Conclusion : In cerebral ischemia-reperfusion, increased serum levels of 1L-18 and COX2 are involved in the death of neuronal cells.
