摘要
(5S)-(4-氟苯)-5-羟基戊酸为制备降血脂药物依替米贝的关键手性中间体。本文综述了制备(5S)-(4-氟苯)-5-羟基戊酸的研究进展。分别从化学法和生物法两方面着重介绍了其不对称合成制备技术的研究现状,并对生物催化制备(5S)-(4-氟苯)-5-羟基戊酸技术的发展进行了展望。
( 5S)-( 4-fluorophenyl)-5-hydroxypentanoic acid is a crucial chiral intermediate for the synthesis of cholesterol-lowering drug Ezetimibe. Progress on synthesis of( 5S)-( 4-fluorophenyl)-5-hydroxypentanoic acid is reviewed. The recent development of asymmetric synthesis of( 5S)-( 4-fluorophenyl)-5-hydroxypentanoic acid by chemical catalysis and biocatalysis are primarily introduced. Meanwhile,further study on biocatalytic preparation of( 5S)-( 4-fluorophenyl)-5-hydroxypentanoic acid is put forward.
