人骨髓和脐带来源间充质干细胞体外分离培养及生物学特性比较 被引量:1

摘要 目的对人骨髓和脐带来源的间充质干细胞(MSC)体外分离培养,P3代细胞表面标志物及分泌细胞因子白细胞介素6(IL-6)的浓度进行比较。方法流式细胞术检测免疫表型,Miliplex试剂盒检测IL-6的浓度。结果两种细胞P3代均高表达CD90、CD105、CD44、CD73)LHLA-ABc抗原,均不表达CD14、CD34、CD45等造血细胞相关抗原,UC-Msc不表达HLA-DR抗原,BM-Msc低表达HLA-DR抗原。骨髓间充质干细胞不同时间点分泌的IL-6浓度均高于脐带间充质干细胞,两者均逐步升高。结论脐带间充质有更低的免疫原性,骨髓间充质干细胞在免疫调控方面更有优势。 AIM To compare in vitro isolation, culture, immune phenotype and the concentrations of interleukin-6 ( IL-6 ) of the third passage of human bone marrow mesenchymal stem cells ( BM-MSC ) and umbilical cord tissue mesenchymal stem cells ( UC-MSC ) .METHODS Flow cytometry and Miliplex kit were used for the determination of phenotype and the concentrations of IL-6. RESULTS Two kinds of cells of P3 generation were high expression of CD90, CD105, CD44, CD73 and HLA-ABC antigens were not expressed CD14, CD34, CD45 and other hematopoietic cell related antigen. UC-MSC did not express HLA-DR antigen, BM-MSC low expression of HLA-DR antigen. BM-MSC at different time points of IL-6 secretion of hormone concentrations were higher in UC-MSC, both of which were gradually increased.CONCLUSION UC-MSC have lower immunogenicity, BM-MSC in the immune regulation has more advantages.
