何首乌中核苷类成分QTRAP UPLC-MS/MS分析

摘要 目的:建立同时测定何首乌中10种核苷类成分的QTRAP UPLC-MS/MS分析方法,分析比较生何首乌和制何首乌中核苷类成分的差异。方法:采用QTRAP UPLC-MS/MS技术,Waters Atlantis T3色谱柱(2.1 mm×150 mm,3μm),流动相甲醇-5 mmol·L-1乙酸铵(含0.1%冰乙酸),0.4 m L·min-1梯度洗脱,采用正离子多反应监测模式测定何首乌商品药材中10种核苷类成分的含量。结果:10种核苷类成分具有良好的线性关系,相关系数均>0.99;何首乌商品药材中核苷类成分尿苷、腺嘌呤、鸟苷、胞苷含量均较高;生何首乌与制何首乌中核苷类成分含量差异明显。结论:该方法简便、灵敏、准确。该研究为何首乌药材内在质量的评价和控制提供可靠的检测方法。 Objective: To develop a UPLC-QTRAP-MS /MS analysis method for the simultaneous determination of 10 nucleosides and nucleobases in Polygoni Multiflori Radix,and analyze the differences of nucleosides and nucleobases in raw and prepared Polygoni Multiflori Radix. Method: QTRAP UPLC-MS / MS technology was performed on a Waters Atlantis T3column( 2. 1 mm × 150 mm,3 μm),eluted by a mobile phase of methanol-5 mmol·L-1ammonium acetate( 0. 1% acetic acid) at a flow rate of 0. 4 m L ·min-1. Ten kinds of nucleosides and nucleobases were analyzed in the positive ion multiple reaction monitoring( MRM) mode. Result:Ten nucleosides and nucleobases had good linearities,correlation coefficient 〉0. 99; the content of uridine,adenine,guanosine and cytidine was higher in Polygoni Multiflori Radix; there was significant difference in nucleoside compositions between raw and prepared Polygoni Multiflori Radix. Conclusion: The method is simple,sensitive,accurate,and can provide a reliable and effective technique for the quality control of Polygoni Multiflori Radix.
