云南松松塔提取物含药血清对乙醇损伤大鼠肝细胞的抗氧化作用 被引量:9

Anti-oxidation Effect of Drug-containing Serum with Pinus yunnanensis on Damaged Rat Hepatocytes Induced by Ethanol

摘要 目的:研究云南松松塔提取物含药血清对乙醇损伤大鼠肝细胞的抗氧化作用进行研究。方法:取SD大鼠10只,随机分为正常组和云南松松塔提取物组(1 g·kg-1),每组5只,每天ig 2次,连续3 d,于末次ig 8 h后制备含药血清。取新生大鼠处死,无菌条件下,取肝脏分离正常肝细胞进行培养,将预培养的肝细胞分为正常组、模型组、空白组、3个不同质量分数云南松松塔提取物含药血清组(25%,50%,100%),用不同质量分数的云南松松塔提取物含药血清对体外培养的大鼠正常肝细胞进行预处理,1 h后,除正常组,空白组外,其余组加入100 mmol·L-1乙醇诱导损伤,继续培养8 h后,MTT法检测大鼠肝细胞存活率,酶标法检测培养液中丙氨酸氨基转移酶(ALT),超氧化物歧化酶(SOD)活性和丙二醛(MDA),还原型谷胱甘肽(GSH)含量。结果:与正常组比较,受损大鼠肝细胞经5%和10%含药血清处理后,存活率明显增大,细胞培养液中ALT活性和MDA含量明显降低,SOD活性和GSH含量明显升高(P<0.05,P<0.01)。结论:云南松松塔提取物含药血清对乙醇损伤大鼠肝细胞有一定的抗氧化作用。 Objective: To study anti-oxidation effect of drug-containing serum with Pinus yunnanensis on the damaged rat hepatocytes induced by ethanol. Method: Ten SD rats were selected and randomly divided into normal group and P. yunnanensis group( 1 g·kg-1),with 5 rats in each group,twice ig daily for continuous 3days,and the drug-containing serum was prepared 8 hours after the last ig medication. The neonatal rats were sacrificed and their normal hepatocytes were separated under sterile conditions for culture. The pre-cultured hepatocytes were divided into normal group,model group,negative group,3 drug-containing serum groups of P. yunnanensis( 25%,50%,100%). The normal hepatocytes were pre-treated with different concentrations ofdrug-containing serum for 1 h. All the other groups except normal group and negative group received ethanol( 100 mmol·L-1) for wound inducing. 8 h after injury,MTT method was used to measure cellular survival rate,and the levels of alanine aminotransferase( ALT),malondialdehyde( MDA),superoxide dismutase( SOD),and glutathione( GSH) in culture medium were measured. Result: Compared with the normal group,the hepatocytes in injured rats had significantly higher survival rate,significantly reduced ALT activity and MDA content in culture medium,and significantly increased SOD activity and GSH content( P〈 0. 05,P 〈0. 01) after treatment with 5%and 10% drug-containing serum. Conclusion: The drug-containing serum with P. yunnanensis has anti-oxidation effect on the damaged rat hepatocytes induced by ethanol.
