透骨草水提物与抗菌药联合用药对耐药大肠杆菌的抑菌效果 被引量:6

Antibacterial effect of aqueous extract from Phryma leptostachaya L. var. asiatica Hara combined with antibiotics on drug-resistant Escherichia coli

摘要 【目的】探讨透骨草水提物与抗菌药联用对产超广谱β-内酰胺酶(ESBLs)大肠杆菌的抑制效果,为扩大透骨草的药用价值及其药源开发利用提供参考依据。【方法】采用二倍微量稀释法和中西药联合诱导传代法测定联合诱导后各代产ESBLs大肠杆菌的最小抑菌浓度(MIC)。【结果】透骨草水提物的MIC为125 mg/m L,以1/2 MIC的透骨草水提物与头孢曲松、头孢他啶、头孢噻呋、头孢噻肟、阿莫西林、磷霉素、粘杆菌素、痢菌净、磺胺间甲氧嘧啶、阿米卡星、氟苯尼考、阿奇霉素、利福平联合诱导传代,至第4代后各抗菌药的MIC降幅均超过4倍;与强力霉素、环丙沙星、诺氟沙星、左旋氧氟沙星、克林沙星、加替沙星联合诱导传代后,其MIC却增加2倍以上。【结论】透骨草水提物与抗菌药联用可通过协同/相加作用或耐药逆转作用而提高抗菌药的抑菌效果,但不宜与氟喹诺酮类抗菌药联合用药。 【Objective】The antibacterial effect of aqueous extract from Phryma leptostachaya L. var. asiatica Hara combined with antibiotics on ESBLs-producing E. coli was investigated, in order to provide references for expanding medicinal value of P. leptostachaya L. var. asiatica Hara and developing its relative medicinal resource. 【Method】The ESBLs-producing E.coli was induced to passage by combining Chinese herbal medicine with Western medicine. Then the MIC of medicines were determined with double microdilution method. 【Result】The result showed that the MIC of P. leptostachaya L. var. asiatica Hara aqueous extract was 125 mg/m L, and the MIC of Ceftriaxone, Ceftazidime, Ceftiofur, Cefotaxime, Amoxicillin, Fosfomycin, Colistin, Mequidox, Sulfamonomethoxine, Amikacin, Florfenicol, Azithromycin, Rifampicin decreased by more than four times since the fourth generation, when combining 1/2 MIC of P. leptostachaya L.var. asiatica Hara aqueous extract with these antibacterial drugs to induce passage of E. coli. However, the MIC of Doxycycline, Ciprofloxacin, Norfloxacin, Levofloxacin, Clinafloxacin, Gatifloxacin increased by more than two times when combining 1/2 MIC of P. leptostachaya L. var. asiatica Hara aqueous extract with these antibacterial drugs to induce passage of E. coli. 【Conclusion 】When combining P. leptostachaya L. var. asiatica Hara aqueous extract with antibacterial drugs, their synergistic/additive effect or reversing effect of drug resistance can improve antibacterial activity of antibacterial drugs, however, it is inadvisable to combine with fluoroquinolones antibiotics.
