寻常型银屑病患者外周血环氧合酶同工酶的研究 被引量:3

Expression of Cyclooxygenase mRNA in Peripheral Blood Mononuclear Cells in Patients with Progressive Psoriasis

摘要 目的探讨进行期斑块状银屑病患者外周血单一核细胞(PBMC)中环氧合酶同工酶的表达及其在银屑病发病机制中的作用。方法应用逆转录-聚合酶链反应(RT-PCR)检测了进行期斑块状银屑病患者及健康对照者各30例PBMC中构成性环氧合酶(COX-1)和诱导性环氧合酶(COX-2)mRNA的表达。结果进行期斑块状银屑病患者PBMC中COX-2mRNA的平均表达水平明显高于健康对照组(P<0.01),且其mRNA的表达水平与其相应的面积及严重度积分(PASI)呈正相关(P<0.01);而两组间COX-1平均表达水平的差异无显著性(P>0.05)。结论进行期斑块状银屑病患者PBMC中COX-2mRNA的表达增高,提示可能通过诱导前列腺素(PGs)的产生参与进行期斑块状银屑病的发病。 Objective To investigate the expression levels of constitutive cyclooxygenase(COX-1)and inducible cyclooxygenase(COX-2)in progressive plaque psoriasis and their clinical significance.Methods Re-verse transcription-polymerase ch ain reaction(RT -PCR)technique was applied to semi-quant itatively analyze COX-1and COX-2mRNA expression in peripheral blood mononuclear cells(PBMC)from 30psoriatic patients and 30healthy controls,and the correlation between COX-1/-2and psoria tic area and severity index(PASI )was evaluated.Results There was a significant difference o f COX-2mRNA expression level betwee n psoriatic pa-tients and normal controls(P<0.01),but no such difference was found reg arding COX-1expression(P>0.05).Apositive correlation was noticed between COX-2and PASI (r=0.68,P<0.01),but not between COX-1and PASI.Conclusion The increased expression of COX-2ca n stimulate over-production of prostaglandins and may be involved in the pathogenesis of pr ogressive plaque psoriasis.[
