腺苷对小型猪脊髓缺血再灌注模型中神经元凋亡及Bcl-2/Bax表达的影响

摘要 目的观察腺苷处理后猪脊髓缺血再灌注实验中神经元凋亡,及其相关蛋白Bax和Bcl-2的表达变化,探讨腺苷对脊髓缺血再灌注的保护作用。方法 24头健康成年西藏小型猪,雌雄不拘,体重20~30 kg,随机分为3组,每组8只:(1)假手术组(n=8):仅游离胸主动脉、锁骨下动脉、副半奇静脉,不阻断血流;(2)缺血再灌注组(n=8):采用胸主动脉、锁骨下动脉阻断法造成脊髓缺血(75 min),在缺血过程中通过副半奇静脉逆行灌注乳酸林格纳液1 000 m L,灌注流速为860 m L/h;(3)腺苷处理组(n=8):手术过程同缺血再灌注组,术中通过副半奇静脉逆行灌注腺苷溶液(1 mg/m L)1 000 m L,灌注流速为860 m L/h。术后动物被放回饲养笼继续饲养,采用Tarlov下肢神经功能评分系统评估6、24、48 h猪的神经运动功能,再灌注48 h各组分别处死8只实验动物,迅速取出来L2~L5及骶段脊髓进行HE、NISSL染色,并采用免疫组化法检测凋亡蛋白Bax和凋亡抑制蛋白Bcl-2的表达,图像分析对结果进行半定量分析。结果 HE及NISSL染色显示假手术组神经元结构完整,腺苷处理组与缺血再灌注组相比神经元病理改变较轻,脊髓神经元凋亡量明显低于缺血再灌注组。免疫组化结果显示假手术组猪脊髓组织中Bcl-2和Bax表达较少,腺苷处理组较缺血再灌注组Bax表达减少和Bcl-2表达增多,差异有统计学意义(P〈0.05)。神经功能评分腺苷处理组术后24、48 h均高于缺血再灌注组,差异有统计学意义(P〈0.05)。结论腺苷处理后可减轻猪脊髓缺血再灌注后神经元凋亡,对脊髓缺血再灌注损伤具有明显的保护作用。其保护作用可能与凋亡蛋白Bax表达减少和凋亡抑制蛋白Bcl-2表达增多有关。
