摘要
为探明线粒体转录因子B2(TFB2 M)基因在贵州白山羊组织中的表达规律,采用实时荧光定量PCR技术测定TFB2 M基因在贵州白山羊心、肝、脾、垂体和下丘脑组织中的表达量。结果表明:目的基因TFB2 M和内参基因β-actin的熔解曲线均呈单一峰形,说明所选条件可准确定量TFB2 M基因在贵州白山羊各组织中的相对表达量。TFB2 M基因在贵州白山羊心、肝、脾、垂体和下丘脑组织中的相对表达量分别为1、0.5092、3.9228、1.9138和0.6744,表现为脾>垂体>心>下丘脑>肝。
The expression quantity of TFB2 Min heart,liver,spleen,hypophysis and hypothalamus issues of Guizhou White Goat was detected by real-time fluorescence quantitative PCR to probe expression pattern of mitochondrial transcription factor B2(TFB2 M)in different tissues of Guizhou White Goat.Results:The melting curves of target gene TFB2 M and reference geneβ-actin both show a single peak,which indicates that the selected condition can exactly quantify the relative expression quantity of TFB2 M in different tissues of Guizhou White Goat.The relative expression quantity of TFB2 Min heart,liver,spleen,hypophysis and hypothalamus tissues of Guizhou White Goat is 1,0.509 2,3.922 8,1.013 8and0.674 4respectively and the relative expression quantity of TFB2 Min different tissues of Guizhou White Goat is spleen〉 hypophysis 〉heart 〉hypothalamus〉liver.
