摘要
目的:探讨加减归肾丸治疗卵巢功能减退的作用机理。方法:SD大鼠随机分为6组,除空白对照组外,予环磷酰胺单剂量腹腔注射造模,分别给予相应药物或生理盐水灌胃4周,各组大鼠每日阴道涂片,观察动情周期变化;给药结束后光镜下观察卵巢组织形态学改变,检测血清FSH、E2。结果:①模型组大鼠动情周期紊乱,中药中、高剂量组动情周期基本正常;②模型组大鼠闭锁卵泡数明显增加,中药高、中剂量组大鼠卵巢形态结构与正常组近似;③中药低剂量组大鼠血清FSH水平明显升高,与空白对照组比较有显著性差异(P〈0.05),中药高及中剂量组、西药对照组血清FSH水平明显降低,与模型组比较有显著性差异(P〈0.05);模型组、中药低剂量组大鼠血清E2水平明显降低,与空白对照组比较有显著性差异(P〈0.05),中药高及中剂量组、西药对照组血清E2水平明显升高,与模型组比较有显著性差异(P〈0.05)。结论:加减归肾丸可恢复DOR大鼠动情周期,促进DOR大鼠卵泡的发育及排卵功能,改善DOR大鼠血清高促性腺激素低雌激素状态,从而恢复卵巢功能。
Objective:To investigate the functional mechanism of Jiajian Shengui Pill treating hypoovarianism. Methods:SD rats were randomly di-vided into six groups, except the blank control group others treated with cyclophosphamide of single dose intraperitoneal injection, respectively with 4 weeks of corresponding drugs or saline irrigation, each group with daily vaginal smear to observe the changes of estrous cycle;after administration under light microscope the morphological changes of ovarian tissue were observed and serum FSH and E2 were detected. Results:①The estrous cy-cle of model group was disorder, estrous cycle of medium and high dose group of Chinese medicine were fundamentally normal;②number of atresia follicles in model group significantly increased, ovarian morphology of medium and high dose group of Chinese medicine is approximate to control group;③serum FSH level of low dose of Chinese medicine group significantly increased, with significant difference of the blank control group (P〈0.05), serum FSH level of medium and high dose group of Chinese medicine and model group of Western medicine significantly decreased, being sig-nificant different with model group (P〈0.05);serum E2 level of model group and low dose group of Chinese medicine significantly decreased, with significant differences with blank control group (P〈0.05), serum E2 level of medium and high dose group of Chinese medicine and control group of Western medicines increased significantly, with significant difference with model group (P〈0.05). Conclusion: Jiajian Shengui pill can restore the emotional cycle of DOR rat, promote the follicular development and ovulation function, improve serum high gonadotrophin low estrogen level of DOR rat, thereby restoring ovarian function.
