摘要
目的:探讨川芎嗪对甲醛所致炎性痛小鼠脊髓背角ERK表达变化的影响,以评价川芎嗪对炎性痛的镇痛效果。方法:将30只昆明种小鼠随机分为正常组(n=6)、生理盐水预处理组(n=12)及川芎嗪(TMP)药物干预组(n=12)。TMP预处理组在注射甲醛之前灌胃TMP(20 g/L),按小鼠体质量0.05 m L/10 g灌胃30 d。生理盐水组小鼠灌胃等量生理盐水。两组均用微量注射器注射0.02 m L 4%甲醛至小鼠右侧脚掌皮下从而建立炎性疼痛模型,通过观察小鼠舔足和咬足时间进行行为学检测,免疫组化荧光检测脊髓背角ERK变化。结果:在疼痛第1时相(注射PF后10 min内),NS预处理组与TMP预处理组两者之间行为学检测比较,差异无统计学意义(P〉0.05);在疼痛第2时相(注射PF后10~45 min),NS预处理组小鼠注射PF后,舔足咬足持续时间为285 s,TMP预处理组注射PF后,舔足咬足持续时间为145 s,两组比较,差异有统计学意义(P=0.0072)。实验小鼠脊髓背角ERK免疫组化荧光检测,TMP预处理组与生理盐水预处理组相应时间点比较,TMP预处理组PF给药30 min、60 min亚组小鼠脊髓背角ERK阳性表达均弱于生理盐水预处理组,差异有统计学意义(P〈0.05)。结论:TMP能调节小鼠背角ERK表达从而调节PF致炎性痛的程度。
Objective: To study the influences of tetramethylprazine on the Spinal dorsal horn ERK expression of inflamma-tory pain mice caused by formaldehyde, in order to evaluate the tetramethylprazine analgesic effect on inflammatory pain. Meth-ods: The 30 Kunming mice were randomly divided into normal group(n=6), Physiological saline preconditioning group(n=12)and tetramethylprazine(TMP) drug intervention group(n =12).TMP preconditioning group before the injection of formaldehyde should be filled the stomach with TMP(20 g/L),and it should be filled the stomach with 30 d according to the weight of mice0.05 ml/10 g. The mice in the physiological saline preconditioning group should be filled the stomach with an equal amount saline. The mice in the TMP drug intervention group and the physiological saline preconditioning group should be injected 0.02 m L 4% formaldehyde to the right side of the sole of the foot in order to set up the inflammatory pain model. We can imple-ment the behavioral detection and immunohistochemical fluorescence detection of spinal cord dorsal horn ERK changes by ob-serving the mice licking and biting time. Results: In the first pain phrase(in 10 minutes after the injection of PF), there is no statistical difference in behavioral detection between NS preconditioning and TMP preconditioning group(P〉 0.05); In the second pain phrase(in 10 to 45 minutes after the injection of PF), the licking and biting foot lasting time of the mice in NS precondi-tioning group can be up to 285 s after injecting PF; the licking and biting foot lasting time of the mice in TMP preconditioning group can be up to 145 s after injecting PF, and there is statistical difference between the two groups( P=0.0072). Laboratory mice spinal cord dorsal horn ERK immunohistochemical fluorescence detection, TMP preconditioning group comparing with saline groups in the corresponding time points, TMP preconditioning group PF for 30 min and 60 min subgroup mice spinal cord dorsal horn ERK positive expression are weaker than the former, so that there is statistical significance( P〈0.05). Conclusion: TMP can adjust mice dorsal horn ERK expression, thus it can regulate the degree of PF inflammatory pain.
