临床耐药性表皮葡萄球菌整合子分子检测分析 被引量:1

Detection and analysis of the prevalence of integrons in clinical drug resistant Staphylococcus epidermidis

摘要 目的了解医院临床耐药性表皮葡萄球菌整合子流行现状,以提高治疗效果。方法对2014年1月-2015年1月的18株临床耐药性表皮葡萄球菌进行回顾性分析,采用PCR方法利用特异性引物进行整合子筛查和分类,用PCR法扩增整合子的可变区,联合测序技术分析整合子携带的耐药基因,分析本地区基因盒的种类。结果 18株临床耐药性表皮葡萄球菌均检出整合子基因,检出率为100%;对整合子阳性菌株整合子进行测序分析,发现18株均为Ⅰ类整合子,对基因盒阳性菌株测序分析显示,基因盒排列为dfr A1+orf C。结论医院感染耐药性表皮葡萄球菌中的整合子主要为Ⅰ类整合子,耐药性表皮葡萄球菌整合子主要携带的是二氢叶酸还原酶耐药基因。 Objective To investigate the current prevalence status of integrons in clinical durg resistant Staphylococcus epidermidis so as to improve the therapeutic effect. Methods From January 2014 to January 2015,a total of 18 strains of Staphylococcus epidermidis were retrospectively analyzed,the screening and classification of integron were carried out by means of PCR. The variable regions of the integron were amplified with the specific primer by PCR,and combined with sequencing technology to analyze the resistance genes of the whole gene,and the types of the local box gene. Results The integron had been detected in18 strains of Staphylococcus epidermidis with the detection rate of 100%; the result of PCR amplification showed that all of 18 strains of Staphylococcus epidermidis were typeⅠintegron. Sequence analysis of gene positive strains showed that the box gene sequence was dfr A1 + orf C. Conclusion The typeⅠintegron is prevalent in the Staphylococcus epidermidis causing nosocomial infections which is related to those strains carry with the dihydrofolate reductase resistance gene.
