摘要
目的确证杭州机场截获发热患者为登革热,为口岸卫生检疫提供技术支持。方法分离患者血清,用ELISA试剂盒检测登革热Ig M抗体;同时提取血清中的病毒核酸,采用登革热通用型实时荧光RT-PCR、RT-LAMP及RTPCR法进行检测,并进行核酸分型检测,同时对RT-PCR的扩增产物进行测序分析。将测序结果与Gen Bank中的其他国家或地区的登革1型病毒株进行比对,分析该病毒的来源。结果血清学检测登革热Ig M抗体为阳性,登革1型病毒RT-LAMP检测结果为阳性,经序列分析该病毒株与印度株(JN940920.1)最为接近,同源性最高,达到97%。结论杭州机场截获的输入性发热患者确证为登革1型病毒感染,该患者为浙江口岸首次发现的输入性登革热。
Objective To confirm the acquisition fever patients from Hangzhou airport as a dengue fever case,and provide technical support for port health quarantine. Methods Serum from the patient was isolated to detect Ig M antibody by ELISA kit; meanwhile,the RNA was extrated and detected by real- time RT- PCR,RT- LAMP and RT- PCR method of dengue virus,and the detection of the case was further typed. The RT- PCR amplification product was analyzed by sequening. The sequencing result and the the other dengue virus type 1 in Gen Bank were compared,analyzing the source of the virus.Results The Ig M antibody and dengue virus type 1 nucleic acids results were positive in the sample. Dengue virus type 1 RT-LAMP test results were positive. Sequence analysis showed that this virus strain was closed to the isolated strains from India( JN940920. 1),with high sequence homology up to 97%. Conclusion The fever patients in Hangzhou airport was confirmed to be infected with dengue virus type 1,which was the imported dengue fever in Zhejiang port.
