定喘止哮颗粒对哮喘豚鼠模型支气管肺泡灌洗液中IL-4和IL-13水平的影响

摘要 目的研究定喘止哮颗粒对哮喘豚鼠模型支气管肺泡灌洗液(BALF)中IL-4、IL-13水平的影响。方法采用10%卵蛋白(OVA)肌肉注射、超声雾化吸入致敏建立支气管哮喘豚鼠模型。造模后随机分为模型组,阳性对照药氨茶碱0.025 g/kg组及定喘止哮颗粒4、2、1 g/kg三个剂量组,另外10只为空白对照组。治疗4周后,观察各组豚鼠支气管肺泡灌洗液中IL-4、IL-13的含量。结果与对照组比较,模型组BALF的IL-4、IL-13明显增加(P<0.01)。定喘止哮颗粒三个剂量组与模型组相比支气管肺泡灌洗液中IL-4、IL-13均有不同程度下降,即定喘止哮颗粒可降低BALF的IL-4、IL-13水平,且剂量越大此作用越明显。结论定喘止哮颗粒可以使IL-4、IL-13水平明显降低(P<0.01),通过调节Th1/Th2细胞之间的功能平衡,达到拮抗气道炎症的作用,且作用强度与药物剂量有关。 Objective To investigate the Effect of Dingchuan Zhixiao granule on the level of IL-4 and IL-13 in bronchoalveolar lavage fluid of the asthma model guinea pig. Methods Guinea pig model of bronchial asthma was established adopting 10% OVA muscle injection and ultrasonic atomizing inhalation. They were randomly divided into model group, The positive control drug aminophylline 0.025 g/kg group,three Dingchuan Zhixiao granule groups(4 g/kg, 2 g/kg and 1 g/kg), and 10 cases in blank control group. After 4 weeks’ treatment, contents of IL-13 and IL-4 in bronchoalveolar lavage fluid were observed. Results Comparimg with control group, the IL-4 and IL-13 in bronchoalveolar lavage fluid of the model group increased significantly(P〈0.01). Comparing with the model group, the IL-4 and IL-13 in bronchoalveolar lavage fluid of the three Dingchuan Zhixiao granule groups had different degrees of decline. Dingchuan Zhixiao granule can reduce the level of IL-13 and IL-4 in bronchoalveolar lavage fluid, and the higher the dose, the more obvious the effect. Conclusion Dingchuan Zhixiao granule can reduce the level of IL-13 and IL-4 in bronchoalveolar lavage fluid significantly, by regulating the function of Th1/Th2 cells, the function of the airway inflammation is reached, and the intensity of action is related to the dose.
