摘要
目的探讨益气活血中药对大鼠退变腰椎间盘中TGF-β1、MMP-3的影响。方法将60只大鼠随机分为对照组(A组)、实验组(B组)、假手术组(C组),每组20只;从腹后外路腰椎间盘纤维环穿刺建立腰椎间盘退变模型,术后第1 d,实验组开始给予益气活血中药浓缩液灌胃,术后第4 w,8 w,每组取5只观察各组椎间盘组织形态学改变,其余5只取标本检测各组退变椎间盘中TGF-β1、MMP-3含量。结果与A组比较,B组大鼠腰椎间盘退行性改变较轻,大鼠腰椎间盘中TGF-β1含量减多(P<0.05)、MMP-3含量减少(P<0.05)。结论益气活血中药可能通过调节TGF-β1、MMP-3的含量,而影响椎间盘细胞外基质合成和降解,来延缓椎间盘退变。
Objective To investigate the effect of benefiting qi for activating blood circulation Chinese medicine on TGF- beta1 and MMP-3in Rats with lumbar intervertebral disc degeneration. Methods 60 rats were randomly divided into control group(group A), the experimental group(group B) and sham group(group C), and each group had 20 cses. Establishing lumbar disc degeneration model from outside of the abdomen. After one day of the operation, the experimental group began administering traditional Chinese medicine and concentrates lavage.After 4weeks and 8 weeks of the operation, five rats were selected in each group, the intervertebral disc tissue morphological changes were observed, and the intervertebral disc degeneration in the TGF-β1, MMP-3 content of the remaining five specimens in each group were detected. Results Comparing with the A group, the lumbar intervertebral disc degeneration of the rats of the B group was lighter, the TGF-β1 content in the lumbar intervertebral disc increased(P 〈 0.05), and the MMP-3 content decreased(P 〈 0.05). Conclusion The benefiting qi for activating blood circulation Chinese medicine may influence the degradation and synthesis of the extracellular matrix through the modulation of TGF-β1 and MMP-3 content, and hence retarded the lumbar intervertebral disc degeneration.
