摘要
目的:探讨microRNA-133b(miR-133b)在食管鳞癌组织中表达及其与临床病理特征的关系.方法:应用实时定量聚合酶链式反应(realtime quantitative PCR)技术检测63例食管鳞癌及其癌旁正常组织中miR-133b的表达量,并分析miR-133b在不同组织中的表达差异及其与食管鳞癌临床病理因素间的关系,探讨miR-133b在食管鳞癌发生与进展中的重要作用.结果:食管鳞癌组织中miR-133b的相对表达量明显低于正常食管钻膜组织(P<0.05),病理分级Ⅲ级和TNM分期Ⅲ期的食管鳞癌患者组织中miR-133b的表达水平明显低于Ⅰ-Ⅱ(P<0.001).结论:miR-133b表达水平与食管鳞癌发生发展与恶性程度密切相关,有望成为食管鳞癌诊断的新靶点.
AIM:To investigate the clinicopathologic significance of microRNA-133 b expression in esophageal squamous cell carcinoma(ESCC).METHODS:Quantitative real-time PCR was used to quantify miR-133 b expression in 63 pairs of ESCC tissues and matched normal esophageal tissues.The association of miR-133 b expression with the clinicopathologic features was then evaluated.RESULTS:The expression of miR-133 b was significantly downregulated in ESCC tissue samples compared to matched normal tissues(P〈0.05).MiR-133 b was highly expressed in ESCC with a low pathological grade(Ⅰ-Ⅱ) but markedly decreased in grade Ⅲ tumors(P〈0.001).In addition,the expression of miR-133 b between patients with TNM stages Ⅰ-Ⅱ and those with stage Ⅲ ESCC was statistically significant(P〈0.001).CONCLUSION:The expression of miR-133 b is closely related to the development and progression of ESCC.MiR-133 b may act as a prognostic marker for this widespread disease.
