摘要
采用紫外分光光度法研究了海带多糖在体外对黄嘌呤氧化酶(XOD)的抑制作用,以氧嗪酸钾作为诱导剂腹腔注射小鼠建立高尿酸血症模型,研究了海带多糖对高尿酸血症小鼠血清尿酸水平和肝脏中XOD的活性的影响。结果表明,体外实验中海带多糖对XOD有明显的抑制作用,其IC50为0.88mg/mL,与别嘌呤醇的联合抑制作用类型为协同作用。海带多糖高、中剂量组均能显著降低高尿酸血症小鼠的血清尿酸水平,并且对肝脏中XOD活性的抑制作用显著。初步认为,海带多糖可以通过抑制XOD的活性对氧嗪酸钾所致的高尿酸血症起到改善作用。
The inhibitory effects of Laminaria japonica polysaccharides on XOD activity were determined by UV-spectrophotometry.Potassium oxonate was injected to induce hyperuricemia in mice.The serum uric acid and the XOD activity in liver were determined.The results showed that Laminaria japonica polysaccharides could inhibit the activity of XOD obviously in vitro,and the IC50 was0.88mg/mL,moreover the study of combined inhibitory effects showed that it was synergy between Laminaria japonica polysaccharides and allopurinol.The Laminaria japonica polysaccharides highdose group and middle-dose group both decreased serum uric acid levels,and the hepatic XOD activity was reduced significantly by Laminaria japonica polysaccharides treatment.It was concluded that Laminaria japonica polysaccharides possesse potent anti-hyperuricemic effects on models of hyperuricemia in mice pretreated with potassium oxonate by inhibiting the activity of XOD.
