活血祛湿方加节食法对大鼠非酒精性脂肪肝模型的影响 被引量:5

Effect of Nonalcoholic Fatty Liver Model Induced by High-fat Purified Diets Fed with the Huoxue Qushi Prescription and Dieting

摘要 目的:观察活血祛湿方灌胃加节食法和单纯节食法对高脂纯化饲料诱导的大鼠非酒精性脂肪肝模型的治疗作用。方法:大鼠随机分为正常组、模型组、节食组、中药加节食组,每组10只,实验周期每周检测各组大鼠体重。正常组予以普通纯化饲料喂养。除正常组外,其余各组均以高脂纯化饲料喂养造模,喂养10周,每组随机处死1只大鼠,观察光镜下肝脏病理切片证明造模成功。模型组继续予以高脂纯化饲料;节食组予以普通纯化饲料喂养;中药加节食组给予普通纯化饲料喂养,并予活血祛湿方提取液按2 g生药/m L的剂量灌胃,连续6周。末次给药后禁食不禁水12 h,予以1%浓度的戊巴比妥按40 mg/kg体重腹腔注射麻醉大鼠,腹主动脉取血,离心取上清,检测甘油三酯(TG)、胆固醇(CHOL)、丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST),摘取肝脏,RT-PCR法测定肝组织中过氧化物酶体增殖物激活受体(PPARα)含量,光镜下观察肝脏病理切片,观察活血祛湿方加节食法对上述指标的影响。结果:与正常组相比,模型组大鼠体重上升明显(P<0.05),TG、CHOL、ALT、AST含量明显上升(P<0.05),肝组织中PPARα含量下降(P<0.05),肝脏病理切片脂肪变更为明显。与模型组相比,节食法和中药加节食法组大鼠体重均有下降(P<0.05),血清中TG、CHOL、ALT、AST水平有不同程度下降(P<0.05),肝组织中PPARα含量增加(P<0.05),肝脏病理切片脂肪变均有所好转。与节食组比较,中药加节食组大鼠体重下降更为明显(P<0.05),血清ALT、AST、TG水平下降更为显著(P<0.05)。光镜下显示中药加节食组在改善肝脏病理切片脂肪变方面比节食组效果更好。两组均可显著提高肝组织PPARα表达(P<0.05),两组间疗效无明显差异(P>0.05)。结论:活血祛湿方加节食法对高脂纯化饲料诱导大鼠非酒精性脂肪肝模型具有治疗作用,优于单纯节食法。其可以降低大鼠体重以及血清TG、CHOL、ALT、AST水平,改善肝脏病理,提高肝组织中PPARα表达可能是其机制。 Objective:To observe the effect of fed with the Huoxue Qushi prescription and dieting on nonalcoholic fatty liver model induced by high-fat purified diets. Methods:Rats were randomly divided into normal group,model group,diet group,TCM plus diet groups of 10,each week detecting the body weight of rats. Normal group were given ordinary purified diet.All the other groups were fed high-fat purification for 10 weeks diet exept the normal group.1 rats in each group were sacrificed each week to prove modeling successfully. The model group continue to be given high-fat diet;the other two groups were given ordinary diet;TCM plus diet group was given ordinary purified diet and the Huoxue Qushi prescription extract at 2 g crude drug/m L for 6 weeks. After the last administration 12 h,intraperitoneal injection pentobarbital to anesthetize rats,getting abdominal aortic blood,centrifuged supernatant,detect triglyceride(TG),cholesterol(CHOL),alanine aminotransferase(ALT),aspartate aminotransferase(AST),the removal of the liver,RT-PCR assay to detect PPARα content of liver tissue,liver biopsy in light microscopy to observe the affect of Huoxue Qushi prescription on these parameters. Results:Compared with normal group,model group rats body weight increased significantly(P〈0.05),TG,CHOL,ALT,AST were significantly increased(P〈0.05),liver tissue PPARα decreased(P〈0.05),liver pathological fat change is obvious. Compared with the model group,rats weight of both groups were decreased(P〈0.05),serum TG,CHOL,ALT,AST levels have declines of different degrees(P〈0.05),liver tissue PPARα content increased(P〈0.05),fatty degeneration of the liver biopsy were improved. Compared with the diet group,body weight of rats in TCM plus diet group decreased more significantly(P〈0.05),serum ALT,AST,TG levels decreased more significantly(P〈0.05). Rats in TCM plus diet group show better improvement in liver biopsy in light microscope than the diet group. Both groups could significantly increase the expression of PPARα in liver tissue(P〈0.05),no significant difference(P〉0.05). Conclusion:TCM plus dieting group has a therapeutic effect on fat purified diet induced non-alcoholic fatty liver model. It can reduce weight and rat serum TG,CHOL,ALT,AST levels,improve liver tissue PPARα expression,improving the treatment of liver pathology so as to achieve the purpose of treatment of nonalcoholic fatty liver.
