摘要
目的:建立腰椎间盘退变的大鼠模型,观察淫羊藿苷对其Ⅱ型胶原蛋白和基质金属蛋白酶表达的影响,从而判断淫羊藿苷是否能改善椎间盘退变。方法:将60只Wistar大鼠随机分为3组:假手术组、对照组、淫羊藿苷组,每组20只。其中对照组、淫羊藿苷组采用纤维环穿刺法建立大鼠椎间盘退变模型,假手术组只做切开,不做穿刺。建立模型后1周,淫羊藿苷组以6.0 g/(kg·d),空白对照组和假手术组予以等量的生理盐水灌胃,持续8周。结果:RT-PCR的方法定量分析各组的Ⅱ型胶原蛋白m RNA和基质金属蛋白酶m RNA的表达。对照组的Ⅱ型胶原蛋白m RNA的表达明显低于假手术组,基质金属蛋白酶m RNA的表达明显高于假手术组。淫羊藿苷组的Ⅱ型胶原蛋白m RNA的表达明显高于对照组,基质金属蛋白酶m RNA的表达明显低于对照组。结论:淫羊藿苷能明显改善椎间盘的退变。
Objective:To observe the effect of ICA type Ⅱ collagen matrix metal protease expression,determine whether ICA can improve disc degeneration. Methods:According to the method of random Numbers,60 Wistar rats are divided into 3 groups:Sham group,Control group and ICA group each 20 only. The Control group and ICA group of the fiber ring puncture method is adopted to establish the rat model of intervertebral disc degeneration,1 week after model,ICA group with 6.0 g/(kg·d) lavage,lasting 8 weeks. Results:RT-PCR analysis of the expression of each groupⅡ type collagen m RNA and MMP m RNA. The m RNA expression of collagen type Ⅱ control group was significantly lower than the sham group,the expression of matrix metalloproteinase m RNA was significantly higher than the sham group. Type Ⅱ collagen m RNA expression of ICA group was significantly higher than the control group,the expression of matrix metalloproteinase m RNA was significantly lower than the control group. Conclusion:ICA can significantly improve the intervertebral disc degeneration.
