摘要
目的 观察低密度脂蛋白(LDL)水平和低密度脂蛋白受体(LDLR)基因表达水平与2型糖尿病(T2DM)的相关性.方法 提取2013年3月~7月就诊于第二炮兵总医院的95例T2DM患者和113例健康对照者外周血白细胞RNA,应用荧光标记技术检测LDLR的基因表达水平;采用全自动生化分析仪及酶法检测血清低密度脂蛋白-胆固醇水平(LDL-C).结果 T2DM组LDLR基因表达水平为1.988±1.158,正常对照组为2.601±2.350,差异有统计学意义(P=0.016);T2DM组LDL-C为2.828±0.806,正常对照组为2.889±0.820,差异无统计学意义(P=0.591),两组的LDLR基因表达和LDL-C水平间无相关性(r=0.070,P=0.503).结论 受体途径可能不是T2DM患者LDL的主要代谢途径,LDLR基因表达水平可作为T2DM的诊断指标.
Objective To investigate the association among low-density lipoprotein (LDL) levels and low-density lipoprotein receptor (LDLR) gene expression levels and type 2 diabetes mellitus (T2DM). Methods Extracted RNA samples of periph- eral blood white cells of 95 patients with T2DM and 113 healthy individuals in March 2013 to July 2013 from the general hospital of the second artillery. LDLR gene expression levels were analysed with fluorescence labeled technology, and the levels of low-density lipoprotein-cholesterol (LDL-C) in serum were detected by automatic biochemical modular system with enzyme. Results LDLR gene expression levels of T2DM group were 1. 988±1. 158, and that of control group were 2. 601 ± 2. 350. It had statistical difference between the two groups (P= 0. 016). The serum LDL-C levels of T2DM group were 2. 828±0. 806 ,and that of control group were 2. 889±0. 820. It had not statistical difference between the two groups (P= 0. 591). The levels of LDLR gene expression of the two groups were not associated with LDL-C (r=0.07,P〉0.05). Con- clusion Receptor method may not be the main metabolic method of LDL in T2DM patients. LDLR gene expression level could be regarded as a diagnostic marker of T2DM.
