摘要
目的通过体外研究观察不同浓度盐霉素及其联合应用紫衫醇、丝裂霉素对膀胱移行细胞癌T24细胞增殖活性的抑制作用。方法将盐霉素(SAL)单药分别配成盐霉素0.1、0.2、0.4、0.8和1.6mg/m L培养液并分不同浓度处理组;盐霉素(SAL)联合紫杉醇(PAC)联合用药处理:使用细胞培养液配成五种不同浓度,以PAC 0.5μg/m L为阳性对照组;盐霉素(SAL)联合丝裂霉素(MMC)联合用药处理五种浓度,并以MMC2.0μg/m L为阳性对照组;各组均设空白及阴性对照。分别于药物处理24h(或48h)及48h(或72h)后通过CCK8法测定吸光度,并计算细胞生长抑制率及相对细胞生长抑制率。结果不同浓度盐霉素单独用药后对细胞的生长出现增殖活性均有抑制作用;不同浓度盐霉素联合紫杉醇对增殖活性明显的抑制作用,且具有一定的时间依赖性。盐霉素联合丝裂霉素对膀胱移行细胞癌T24细胞的体外增殖活性有显著抑制作用。结论不同浓度盐霉素单用及联合应用紫杉醇或丝裂霉素均对T24细胞增殖活性有抑制作用,并且与浓度和时间一定的依赖性。
Objective To observe inhibition effect of various concentration of salinomycin combined with paclitaxel or with mitomycin on bladder transitional cell carcinoma T24 cell proliferative activity in vitro study. MethodsCulture medium with 0.1mg/m L, 0.2mg/m L, 0.4mg/m L, 0.8mg/m L and 1.6mg/m L of salinomycin were respectively prepared and they were allocated to different concentration treatment group. Combined medication of SAL with paclitaxel(PAC) included that cell culture medium was prepared into five different concentration and positive PAC 0.5μg/m L was set as the control group. Combined medication of SAL with mitomycin(MMC) was prepared into five different concentration and positive MMC 2.0μg/m L set as the control group. Each groupHad bank control and positive control. Absorbance of 24 hours(or 48 hours) and 48 hours(or 72 hours) after medicament treatment was detected by CCK8 method. In addition, cell growth inhibition rate and relative cell growth inhibition rate were calculated. Results Different concentration of SAL had different inhibition effects on cell proliferative activity. Different concentration of SAL combined with PAC had obvious inhibition effect on cell proliferative activity and it had time-dependent manner. SAL combined with MMC had significant in vitro inhibition effect on bladder transitional cell carcinoma T24 cell proliferative activity. Conclusion Single mediation of SAL or different concentration of SAL combined with PAC or MMC all had inhibition effect on T24 cell proliferative activity and it is dependent on concentration and time to some degree.
