急性冠脉综合征患者外周血单核细胞miR-145与超敏C反应蛋白的相关性研究 被引量:4

Study of Correlation between Peripheral Blood Mononuclear Cells miR-145 and High-sensitivity C-reactive Protein in Patients with Acute Coronary Syndrome

摘要 目的探讨急性冠脉综合征(ACS)患者外周血单核细胞(PBMCs)miR-145与超敏C反应蛋白的相关性。方法选取急性冠脉综合征患者90例,其中不稳定型心绞痛(UAP)30例(A组),ST段抬高型心肌梗死(STEMI)30例(B组)和非ST段抬高型心肌梗死(NSTEMI)30例(C组)。另外30例以心前区不适住院,既往无高血压、糖尿病和冠心病史,经心电图、心肌酶学和冠状动脉造影排除冠心病者作为对照组(D组)。采用Real-time PCR法检测PBMCs中miR-145表达水平,使用U6作为内参对照。采用ELISA法检测超敏C反应蛋白(hs-CRP)水平。结果 A组、B组、C组分别与D组比较,ACS患者外周血单核细胞miR-145表达水平明显降低,均有统计学意义(P<0.05);A组、B组、C组分别与D组比较,PBMCs培养基上清中细胞因子hs-CRP水平明显升高,均有统计学意义(P<0.05);Pearson相关分析结果显示,ACS患者外周血单核细胞miR-145表达与hs-CRP水平呈负相关性(r=-0.84,P<0.05)。结论 ACS患者外周血单核细胞miR-145与超敏C反应蛋白之间呈负相关性,可能成为ACS新型标志物。 Objective To investigate correlation between the peripheral blood mononuclear cells miR-145 and high-sensitivity Creactive protein in patients with acute coronary syndrome. Methods 90 patients with ACS were randomly selected. 30 were diagnosed as unstable angina pectoris( UAP,group A),30 patients were diagnosed as STEMI( group B) and 30 patients were diagnosed as NSTEMI( group C). Another 30 cases were selected as a control group( group D) hospitalized with chest discomfort,with no previous history of hypertension,diabetes and coronary heart disease,and excluding coronary heart disease by the electrocardiogram,myocardial enzymes and coronary angiography. Using Real-time PCR to detect the expression level of miR-145 in PBMCs,while using U 6 as an internal reference control. Hs-CRP levels were detected by ELISA. Results group A,group B,C were compared with group D respectively,expression level of peripheral blood mononuclear cells miR-145 in patients with acute coronary syndrome was significantly lower( P 0. 05); levels of hs-CRP of supernatant cytokine cultured in PBMCs were significantly elevated( P 0. 05); pearson correlation analysis showed that expression levels of peripheral blood mononuclear cellsmiR-145 and hs-CRP were correlated negatively inpatients with ACS( r = 0. 84,P 0. 05). Conclusions Peripheral blood mononuclear cellsmiR-145 and hs-CRP were correlated negatively in patients with acute coronary syndrome,that may become a new biomarker for acute coronary syndrome.
