摘要
目的研究哇巴因对大鼠胸主动脉血管平滑肌细胞(vascular smooth muscle cells,VSMCs)中纤维连结蛋白(fibronectin,FN)表达的影响,探讨钠泵α亚单位离子转运活性在ECM形成中的作用。方法采用组织块贴壁法进行大鼠胸主动脉VSMCs原代培养,并运用免疫组织化学方法进行细胞鉴定并检测FN在VSMCs中的表达。结果随着培养液内哇巴因浓度的增加,VSMCs中FN的表达逐渐下调,而且半定量结果分析显示,不同剂量哇巴因干预组间VSMCs中FN的表达存在统计学差异。结论哇巴因可能通过抑制VSMCs钠泵活性而下调FN的表达,为高血压的研究提供新的实验依据。
Objective To investigate the effect of ouabain on the expression of fibronectin( FN) in rat thoracic aorta vascular smooth muscle cells( VSMCs),and to explore the role of the sodium pump alpha subunit ion transport activity in extracellular matrix( ECM)formation. Methods Primary thoracic aorta VSMCs from SD rats were cultured by tissue adherent method,The expression of FN was detected in VSMCs. Results The expression of FN was decreased gradually with the increasing of ouabain concentration,and there was significant difference after treated with different concentrations of ouabain. Conclusion Ouabain can down-regulate the expression of FN in rat vascular smooth muscle cells by inhibiting the activity of Na^+-K^+-ATPase,which provides a scientific experimental evidence for the pathogenic mechanism of hypertension.
