摘要
利用柱层析法对玉米胚芽毛油中酯态甾醇和游离态甾醇进行分离,并分别用正己烷/乙酸乙酯(体积比为90∶10)和正己烷/乙醚/无水乙醇(体积比为25∶25∶50)洗脱,之后将这两种组分分别进行皂化、提取、衍生化并利用气相色谱法定量.此方法测得酯态甾醇和游离态甾醇的相对标准偏差分别为1.32%、1.46%,精密度高,稳定性好,并加入实验室自制β-谷甾醇亚油酸酯作为酯态甾醇标准品,回收率分别为90.71%、94.73%,可用于玉米胚芽油中两态甾醇的分离以及含量的测定.
Plant sterols have the important physiological functions, such as reducing serum low density lipoprotein (LDL), antioxidant, anti-inflammatory, anticancer, antibacterial and so on. They always exist in the forms of sterol ester (SE), hydroxyl sterol ester (HSE), steroidal glycosides (SG)and acyl steroidal glycosides (ASG)in the plant oils. The present study was to examine the separation and analysis methods of the esterified and free sterols in the corn germ oil. The results showed that the esterified and free sterols were separated by silica gel column chromatography upon elution with n-hexane/ethyl acetate (90:10)followed by n-hexane/diethyl ether/ethanol (25:25:50). Both fractions were saponified, extracted, and derivatized and the phytosterol content was quantified by GC. This analytical method for the esterified and free sterols had a relative standard deviation of 1.32% and 1.46% ,respectively and the recovery rates were 90.71% and 94.73% ,respectively,while the self-made α-sitosterol linoleic acid ester as internal standard. It was concluded that this analytical method had high precision and good stability and could be used for the separation and determine of the two states of phytosterols in the corn germ oil.
