摘要
采用微波协同纤维素酶法提取薯蓣(Dioscorea opposita)多糖,并利用苯酚-硫酸法测定多糖的含量,在单因素试验的基础上,通过正交试验选出优化的组合因素。结果表明,最佳提取工艺条件为纤维素酶添加量0.20%、酶解时间70 min、酶解温度45℃、pH 5.0、微波功率600 W、微波时间50 s,在此条件下,薯蓣多糖提取含量为30.25 mg/g,明显高于其他提取方法。
Adopting benzene polyphenol-hydrous method to determine the content of polysaccharides, the optimum extraction technology of Dioscorea opposita polysaccharide with cellulase enzymolysis under microwave was studied through single factor and orthogonal experiment. The results showed that, the best extraction process was cellulase enzyme quantity 0.2%, extract-ing time 70 min, extraction temperature 45℃, pH 5.0, microwave output power 600 W and microwave radiation time 50 s. The extraction quantity of D. opposita polysaccharide reached to 30.25 mg/g under this condition.
