摘要
目的研究侧脑室注射Apelin-13对脑缺血再灌注损伤大鼠神经缺损程度及脑内(Yes—asso—ciatedprotein,YAP)表达的影响。方法健康成年Wistar雄性大鼠,采用双盲法随机分为对照组、脑缺血一再灌注组和Apelin-13干预组,每组3只大鼠。给予大鼠禁食禁水12h后,采用线栓法制作大鼠脑缺血一再灌注模型,缺血1h,再灌注24h,Apelin-13干预组于再灌注后经侧脑室注射Apelin-13。检测大鼠神经功能缺损程度进行行为学评定,取材进行TTc染色检测大鼠脑梗死体积,蛋白免疫印迹法检测大鼠脑组织各部位YAP的表达变化。结果与脑缺血再灌注组相比[(2.67±0.33)分],Apelin-13干预组大鼠神经缺损评分[(1.67±0.33)分]有所降低,症状减轻,差异有统计学意义(P〈0.05),脑梗死体积减小[(30.60±1.42)%vs(23.70±2.20)%,P〈0.05],脑组织各部位YAP表达均上调,差异有统计学意义(P〈0.05)。结论Apelin-13通过调节YAP的表达在大鼠脑缺血一再灌注损伤中发挥治疗效果,具有一定的神经保护作用。
Objective To investigate the neuroprotective effect of lateral intracerebroventricular injection of Apelin-13 on the YAP expression and the cerebral ische mia/reperfusion (I/R) injury in Wistar rats. Methods Healthy adult male Wistar rats were randomly divided into the sham group, cerebral I/R group and Apelin-13 treatment group. The middle cerebral artery occlusion model was established with ischemia for 1 hour and reperfusion for 24 hours after restricting food and water intake for 12 hours. Apelin-13 was injected into rats" lateral ventricles of Apelin-13 treatment group after reperfusion. Neural function defects was assessed. The volume of infarction was evaluated by TFC staining. The expression levels of YAP were detected by western blot. Results Compared with the cerebral I/R group,the rats in the Apelin-13 treatment group had abetter neurologic score ( ( 2.67±0.33 ) vs ( 1.67±0.33 ) , P〈0.05 ) , the infarction volume was decreased ( ( 30.60± 1.42 ) % vs (23.70±2.20) %, P〈0.05 ) , and YAP expression level was increased in each part of the cerebral tissue(P〈0.05). Conclusion Apelin-13 has a neuroprotective effect, which plays the therapeutic effect by regulating the expression of YAP on cerebral isehenfia/ reperfusion (I/R) injury in Wistar rats.
