摘要
目的:研究胃癌组织中蛋白磷酸酶2A抑制剂-2(I2PP2A)基因表达量及基因沉默对胃癌细胞恶性生物学的影响。方法:采集胃癌患者的胃癌组织及癌旁正常组织,培养胃癌细胞株BGC-823并转染I2PP2A的siRNA,检测组织标本中I2PP2A的mRNA含量和蛋白含量;检测细胞样本中的凋亡细胞比例、不同细胞周期比例、Cyclin B2、Cyclin D1、CDK1、Bcl-2、Survivin的蛋白含量。结果:胃癌组织中I2PP2A的mRNA含量和蛋白含量均高于癌旁正常组织;转染I2PP2A-siRNA后,胃癌细胞的G0/G1期细胞比例、凋亡细胞比例高于阴性对照组,S期细胞和G2/M期细胞比例低于阴性对照组,Cyclin B2、Cyclin D1、CDK1、Bcl-2、Survivin的蛋白含量均低于阴性对照组。结论:胃癌组织中I2PP2A基因的表达量异常增多;沉默I2PP2A基因能够通过抑制Cyclin B2、Cyclin D1、CDK1、Bcl-2、Survivin的表达来诱导胃癌细胞凋亡并使细胞周期停滞于G0/G1期。
Objective:To study the expression of I2PP2 Agene in gastric cancer tissues and the effects of gene silencing on malignant biology of gastric cancer cells.Methods:Gastric cancer tissues and adjacent normal tissue in patients with gastric cancer were collected,gastric cancer cell lines BGC-823 were cultured and I2PP2 AsiRNA was transfected.The mRNA contents and protein contents of I2PP2 Ain tissue samples were detected;the proportions of apoptotic cells and different cell cycles as well as protein contents of Cyclin B2,Cyclin D1,CDK1,Bcl-2and Survivin in cell samples were detected.Results:mRNA contents and protein contents of I2PP2 Ain gastric cancer tissues were higher than those in adjacent normal tissues.After transfection of I2PP2A-siRNA,the proportions of G0/G1 phase cells and apoptotic cells in gastric cancer cells were higher than those in negative control group,proportions of S-phase cells and G2/M phase cells were lower than those in negative control group,and protein contents of Cyclin B2,Cyclin D1,CDK1,Bcl-2and Survivin were lower than those in negative control group.Conclusion:The expression of I2PP2 Ain gastric cancer tissue abnormally increases;silencing I2PP2 Agene can inhibit the expression of Cyclin B2,Cyclin D1,CDK1,Bcl-2and Survivin to induce gastric cancer cell apoptosis and make cell cycle stagnate in G0/G1 phase.
