猪Viperin主要抗原区的原核表达及多克隆抗体的制备

摘要 采用PCR方法扩增猪Viperin蛋白主要抗原区基因,克隆入原核表达载体p ET32a,转化大肠杆菌BL21构建重组表达菌,经IPTG诱导表达重组蛋白,SDS-PAGE鉴定蛋白得到表达,分子质量约为40 ku。进一步优化蛋白表达条件为:IPTG终浓度为0.6 mmol/L,诱导5 h。采用His标签单抗对重组蛋白进行Western blot鉴定表明蛋白具有良好的抗原性。将蛋白质纯化后免疫新西兰大白兔,间隔14 d免疫4次,并采集血清。用间接ELISA方法测定血清抗体效价,结果表明,经过4次免疫血清抗体效价达到1∶409600,间接免疫荧光检测出现特异荧光,表明其能与Viperin蛋白发生反应。本试验为研究Viperin的功能、建立特异的检测方法奠定了基础。
