Association of Clock Gene rs11133391 Polymorphism with Depressions and Primary Insomnia

摘要 目的:对clock基因的rs11133391多态性位点进行分析并探讨其与抑郁症患者、原发性失眠患者的关系。方法:采用美国ABI公司的小测序SNP分型技术对155例重性抑郁症患者、105例原发性失眠患者和150名正常对照进行clock基因rs11133391位点分型,对clock基因的rs11133391的基因型和等位基因的频率进行比较。结果:抑郁症患者、原发性失眠患者与正常对照之间clock基因rs11133391多态性的等位基因与基因型没有显著差异(P>0.05);与对照组相比,原发性失眠患者rs11133391多态性的等位基因频率有显著差异(P<0.05);对rs11133391多态性三种基因型C/C、C/T、T/T抑郁症和原发性失眠的临床资料比较,显示原发性失眠患者的三种基因型之间有差异(P<0.05),其他各项没有明显差异(P>0.05)。结论:clock基因rs11133391多态性可能与抑郁症的发病无关联,但与原发性失眠可能关联,且与原发性失眠患者的年龄有关。 Objective: To explore the association of clock gene with depression and Primary insomnia. Methods: The clock gene rs11133391 was genotyped by Snapshot SNP technique in 155 patients with major depression, 105 patients with Primary insomnia and 150 healthy controls. We compared the genotype and allele gene frequency in three groups. Results:There were no significant differences in genotype and allele gene in clock gene rs11133391 between depression patients and healthy controls(P〈0.05); There were significant differences in allele gene in clock gene rs11133391 between Primary insomnia and healthy controls(P〈0.05); There were no significant differences of clinical characteristics between three genotypes of the rs11133391 polymorphism C/C、C /T and T/T in depression patients, while in patients with Primary insomnia,significant differences in the clinical symptoms and age were found between three genotypes of the rs11133391 polymorphism. Conclusion: Clock gene rs11133391 polymorphism may have associations with Primary insomnia, but not depression.
