Application of LC- MS- based metabolomics in the evaluation of renal toxicity and identification of biomarker

摘要 肾毒性作为一种临床常见的毒副作用,导致机体氨基酸代谢和脂类代谢的异常。为此,肾毒性作用机制以及疾病诊断、药物治疗的相关性研究已受到越来越多的关注。在肾毒性研究与评价中,基于液相色谱-质谱(LC-MS)的代谢组学技术已应用于评价肾毒性和鉴定生物标示物,并且快速而准确地预测和诊断肾毒性。在查阅国内文献3篇和国外文献44篇的基础上(1999-2014),本文总结并分析了基于LC-MS的代谢组学技术在化学药物、中药、化学试剂、重金属等诱导的肾毒性研究中的应用进展。 Renal toxicity is a common clinical side effect,which can cause dysregulation of amino acid metabolism and lipid metabolism. Therefore,the action mechanism,disease diagnosis and drug treatment of renal toxicity have been attracting more and more attention. Liquid chromatography- mass spectrometry( LC- MS)- based metabolomics has been well accepted as a powerful tool in renal toxicity study. LC- MS- based metabolomics has also been employed to evaluate renal toxicity and discover biomarkers. This review provides a summary of the reported articles including three articles in Chinese and forty- four articles in English from 1999 to 2014,and presents an overview of LC- MS- based metabolomics in renal toxicity induced by chemical drugs,traditional Chinese medicine,chemicals and heavy metal.
