摘要
以欧洲黑杨N46无性系(Populus nigra‘N46’)的腋芽无菌萌发的幼嫩叶片为试验材料,对影响其不定芽分化和生根的关键因子进行了研究,建立了高效稳定的不定芽诱导和植株再生体系。结果表明:欧洲黑杨N46无性系幼嫩叶片进行不定芽分化诱导的最适培养基为MS+6-BA 0.5 mg·L-1+NAA0.03 mg·L-1+蔗糖30 g·L-1+琼脂粉5 g·L-1,诱导最佳温度为27℃,每个叶片平均分化不定芽数量可达45.20个;不定芽生根的最佳培养基为1/2MS+IBA0.01 mg·L-1+NAA0.01 mg·L-1+蔗糖30 g·L-1+琼脂粉5 g·L-1,生根最佳温度为25℃,生根率可达93.33%,生根小植株移栽成活率达98%。
Using young leaves germinating from the in vitro axillary bud in Populus nigra clone‘N46'from Russia,we studied the key factors affecting the adventitious bud differentiation and rooting,and built a high efficient and stable adventitious bud induction and regeneration system. The best differentiation medium of P. nigra‘N46'was MS basic medium supplemented with 0. 5 mg·L^- 16- BA and 0. 03 mg·L^- 1NAA,and the rooting medium was 1 /2MS supplemented with 0. 01 mg·L^- 1IBA and 0. 01 mg·L^- 1NAA. The suitable temperature for differentiation and rooting was 27 ℃ and 25 ℃,respectively. The totally buds generated from each leaf were 45. 20 on average. The rooting rate of the regenerated plants was 93.33%,and the survival rate in transplanting was 98%.
