摘要 目的研究Drosha蛋白在胃腺癌发展进程中的意义及与患者预后的相关性,探究Drosha对胃癌细胞SGC-7901侵袭能力的影响。方法免疫组织化学染色及Image-Pro Plus软件检测组织芯片889例胃腺癌组织样本中Drosha的表达量,统计分析Drosha与各临床参数及309例胃腺癌患者预后的关系;小干扰RNA(siRNA)干扰人胃腺癌细胞株SGC-7901中Drosha的表达,TranswellTM侵袭实验检测细胞侵袭能力。结果 Drosha在高分化胃腺癌组织中高表达(吸光度值中位值为0.4195),在中分化胃腺癌中表达降低、在低分化腺癌中低表达;Drosha的表达与肿瘤大小、Lauren分型、肿瘤浸润深度、淋巴结转移、肿瘤病理分级及病理分期相关;生存分析显示Drosha高表达的患者拥有更高的生存率;敲低SGC-7901细胞的Drosha水平后,细胞侵袭能力显著增强。结论 Drosha蛋白水平随胃腺癌分化程度降低而逐渐降低,敲低Drosha水平可促进胃癌细胞的侵袭。 Objective To investigate the role of Drosha expression in the progression of gastric adenocarcinoma and its impact on the invasive ability of SGC-7901 human gastric cancer ceils. Methods Drosha expression was detected in 889 gastdc carcinoma samples on tissue microarrays by immunohistochemistry staining and quantified by Image-Pro Plus software. Statistical analysis was used to evaluate the correlations between Drosha expression and the clinicopathological characteristics of the 889 tumor cases or the outcomes of 309 gastric adenocarcinoma patients. Drosha was knocked down in SGC-7901 cells by small interfering RNA (siRNA), and cell invasive ability was assessed by TranswellTM assay. Results Drosha expression was the highest in the well differentiated gastric adenocarcinoma (median absorbance, 0.4195), and the lowest in the poorly differentiated samples. Drosha expression was significantly related to Laren classification, tumor size, tumor invasion depth, lymph node metastasis, tumor pathological grade and stage. Patients in Drosha positive group had a higher survival rate than those in Drosha negative group. Silencing Drosha in SGC-7901 cells resulted in an enhanced cell invasion. Conclusion Drosha expression was reduced gradually with the degrading histological differentiation of gastric adenocarcinoma, and the knock-down of Drosha expression could promote gastric adenocarcinoma cell invasion.
