摘要
目的:体内外研究新型近红外荧光染料IR-808对肿瘤的检测作用,并初步探索其潜在的临床应用。方法:体外培养3种人肿瘤细胞(结肠癌细胞系HCT116、肺癌细胞系A549和乳腺癌细胞系MCF-7),与浓度为20μmol/L的IR-808共孵育20 min,以正常人胚肾细胞系293T作为对照;建立乳腺癌皮下瘤动物模型,静脉注射剂量为1 mg/kg的IR-808,24 h后观察体内成像效果;收集1例恶性胸水标本,分别与同等浓度的染料孵育24和48 h,观察其荧光信号的稳定性,并与临床病理结果比较,判断其检测肿瘤细胞的效果。结果:体内外实验证明近红外荧光染料IR-808只靶向肿瘤成像;与胸水共孵育后,IR-808信号稳定,初步证实其能检测到标本中的肿瘤细胞。结论:IR-808具有良好的靶向肿瘤成像的性能,利用此性质可以用来检测临床标本中的肿瘤细胞,具有重要的应用前景。
Objective To explore IR-808 near-infrared fluorescence dye for its in vivo and in vitro application to tumor detection and other clinical application. Methods Human malignant cell lines(colon cancer cell line HCT116, lung cancer cell line A549 and breast cancer cell line MCF-7) were incubated with IR-808 with a concentration of 20 μmol/L for20 min, and human normal cells embryonic kidney cells(293T) were used as control. The subcutaneous tumor xenografts models were established by bearing human breast cancer cells, the imaging effect was observed 24 h after i.v injection of IR-808 at the dose of 1 mg/kg. One malignant pleuroperitoneal effusion sample was collected and incubated with IR-808 at the same concentration 24 and 48 h after, and then the stability of fluorescence signal and the tumor imaging effect were detected subsequently. Results The targeted imaging performance to cancer cells using IR-808 was confirmed in vitro and in vivo, and the fluorescence signal was very stable. Cancer cells in clinical effusion incubation with IR-808 were also detected. Conclusion IR-808 exhibits the property of tumor targeted imaging, which could be further used to detect tumor cells in clinical samples. So IR-808 will have great potential application in the future.
